摘要
通过腹腔注射CCl_(4)诱导建立小鼠肝损伤模型,研究了口服泥鳅皮冻干粉(LPLS)对肝损伤小鼠血清中肝功能标志物、肝组织中氧化应激标志物水平、肝组织的病理学、肝细胞凋亡水平、肝组织中抗氧化应激和细胞凋亡相关信号通路主要参与者mRNA与蛋白表达变化的影响。研究发现,口服LPLS能以剂量依赖的方式,显著改善CCl_(4)引起的小鼠肝组织病理性变化和肝细胞凋亡程度,显著降低肝损伤小鼠血清中天冬氨酸转氨酶(AST)和丙氨酸转氨酶(ALT)的活性,降低肝组织中丙二醛(MDA)含量,增加超氧化物歧化酶(SOD)活性和还原型谷胱甘肽(GSH)含量;在mRNA和蛋白水平分别上调其核红细胞2相关因子2(Nrf2)、血红素加氧酶1(HO-1)、B淋巴细胞瘤-2(Bcl-2),下调其kelch样ECH相关蛋白1(Keap-1)和Bcl-2相关X蛋白(Bax)的表达。因此,LPLS可通过调节氧化应激、肝细胞凋亡发挥肝脏保护作用。
Mouse liver injury model was induced by intraperitoneal injection of CCl_(4).The effects of oral administration of lyophilized powder of loach skin(LPLS)on the liver function biomarkers in serum,oxidative stress biomarkers in tissue,histopathology changes,the hepatocyte apoptosis level,and the mRNA and protein expression of main factors involving in anti-oxidative stress or apoptosis pathways in liver tissue were studied.This study revealed that LPLS intake significantly improved the histopathologic modification and hepatocyte apoptosis caused by CCl_(4) in a dose-dependent manner,also significantly reduced the levels of alanine transaminase(ALT)and aspartate transaminase(AST),decreased the level of malondialdehyde(MDA),as well as increased the activity of superoxide dismutase(SOD)and the content of glutathione(GSH)in liver.Furthermore,LPLS upregulated the expression of nuclear factor erythroid 2-related factor 2(Nrf2),heme oxygenase-1(HO-1),B-cell lymphoma-2(Bcl-2)and downregulated the expression of kelch-like ECH-associated protein 1(Keap-1)and Bcl-2-associated X(Bax)on both mRNA and protein level.In short,LPLS can protect the liver by regulating both of oxidative stress and hepatocyte apoptosis in vivo.
