转录因子EB调控机制及其对血液肿瘤的影响及靶向治疗研究现状

摘要 转录因子(TF)EB是小眼畸形(MiT)/TFE家族的成员之一,参与调控机体多种生理、病理过程,其活性及亚细胞定位受到多个环节的调控,如转录调控、转录后调控、翻译水平调控及翻译后修饰(PTM)等。TFEB可以以自噬溶酶体依赖或非依赖途径影响多种血液肿瘤的发生、发展,TFEB的靶向调控已成为血液肿瘤的治疗策略之一。笔者就近年来TFEB调控机制及其对血液肿瘤的影响及靶向治疗方法的研究现状进行阐述,旨在为血液肿瘤治疗的潜在靶点选择提供参考。 Transcription factor(TF)EB,a member of microphthalmia(MiT)/TFE family,is involved in regulation of various physiological and pathological processes.Its activity and subcellular localization are regulated by multiple links,such as transcriptional regulation,post-transcriptional regulation,translational level regulation,and post-translational modification(PTM),etc..TFEB can affect occurrence and development of a variety of hematological neoplasms through autophagolysosomal dependent or independent pathways,and targeted regulation of TFEB has been successfully used as a therapeutic strategy for hematological neoplasms.In this article,the current status of research on regulatory mechanisms of TFEB,its impact on hematological neoplasms,and targeted therapeutic approaches in recent years are elaborated,with aim of providing a reference for selection of potential targets for hematological neoplasms therapy.
