摘要
目的:构建并验证血管生成相关基因作为评价肝细胞癌(HCC)患者预后的模型。方法:首先利用TCGA-LIHC数据集进行基因差异表达分析,并基于差异基因构建共表达网络,筛选与HCC增殖、转移及血管侵袭相关的模块,将模块基因与血管生成相关基因取交集,最后通过单因素及多因素Cox回归分析构建预后模型,并进一步对该模型性能及其临床相关性进行评价。ICGC数据作为验证集进行外部验证。结果:筛选出8个血管生成相关基因用于构建预后模型,可作为HCC患者的独立预后因素,风险评分越高的患者生存时间越短,1、2、3年的ROC曲线下面积(AUC)分别为0.797、0.743、0.711。结论:该模型对HCC患者的预后有较好的预测能力,有一定的临床参考价值,同时为HCC血管生成机制研究提供了新的思路。
Objective:To construct and validate a prognostic model of angiogenesis-related genes in hepatocellular carcinoma(HCC).Methods:Differentially expressed genes were obtained by differential analysis of the TCGA-LIHC dataset.Then the co-expression network was constructed based on them to screen the modules related to HCC's proliferation,metastasis,and vascular invasion.Then module genes and angiogenesis-related genes were intersected to obtain candidate genes.Finally,the prognostic models were constructed by univariate and multivariate Cox regression.And further,the performance of the model and its clinical relevance was evaluated.ICGC data were used for validation.Results:Eight genes related to angiogenesis were selected to construct the prognostic model.The ROC curve showed that patients with higher risk scores had shorter survival times.The area under the curve(AUC)of 1,2,and 3 years were 0.797,0.743,and 0.711,respectively,and it indicated that the model could be used for predicting independent prognostic factors for HCC.Conclusion:Our prognostic model can predict the prognosis of HCC patients,which can be used for clinical reference and provide a new idea for the angiogenesis mechanism in HCC.
