摘要
在使用体外培养的细胞进行药物实验时,药物处理会造成细胞死亡,而检测细胞存活率的方法不同,会对实验结果造成影响。以海拉细胞作为实验对象,用不同浓度的顺铂处理细胞,做去除以及不去除旧培养基的处理,比较包含或不包含旧培养基中漂浮细胞情况下检测得到的细胞存活率。实验结果显示,使用CCK8法检测96孔板、24孔板细胞存活率,不去除旧培养基时实验组细胞存活率高于去除旧培养基实验组细胞存活率。在顺铂浓度为0.625μM时,不去除旧培养基与去除旧培养基实验组之间的细胞存活率有显著差异。在6孔板和100mm培养皿中,使用细胞计数仪对用药后的细胞培养基中的漂浮细胞进行检测,发现培养基中漂浮的细胞中存在活细胞。结果表明药物处理后的培养基中存在漂浮的活细胞,去除旧培养基会去除这部分活细胞,从而影响细胞药物实验结果的准确性。
When conducting drug experiments on cells cultured in vitro,drug treatment can cause cell death,and different methods for detecting cell survival rates can impact the experimental results.This study uses HeLa cells as the experimental object and treats the cells with different concentrations of cisplatin.The cell survival rates detected with or without floating cells in the old culture medium are compared.The experimental results indicate that when using the CCK-8 method to detect the cell survival rates in 96-well and 24-well plates,the cell survival rates of the experimental group without removing the old culture medium are higher than those of the experimental group with the old culture medium removed.At a cisplatin concentration of 0.625μM,there is a significant difference in cell survival rates between the experimental groups with and without the removal of the old medium.In 6-well plates and 100 mm dishes,a cell counter is used to detect the floating cells in the cell culture medium after drug application,and the presence of viable cells among the floating cells in the medium is found.The results suggest that the floating viable cells in the culture medium after drug treatment can affect the accuracy of the cell drug experiment results,and removing the old culture medium will remove these viable cells.
