白及不同地理群体的分子遗传学评价

Molecular Genetic Evaluation of Different Geographical Populations of Bletilla striata Rchb.f.

以采自6个不同地理区域33份白及资源为试材,利用SRAP标记技术对白及资源进行分子遗传学评价研究,以期为白及品种资源的综合利用、野生资源的就地保护和迁地保护提供参考依据。结果表明:白及资源在物种水平遗传多样性比较丰富,白及居群的遗传多样性指数分别为Ht=0.3267,Hs=0.2356,Gst=0.2506,Nm=1.4952,说明白及居群间存在少量的基因交流,而且遗传变异主要发生在居群内。6个野生居群根据UPGMA聚类,可分为2个支系。居群结构分析结果表明,K=2时,样品分类效果最佳。Mantel test检测表明,白及居群间遗传距离越大居群之间地理距离就越远,呈显著正相关性(r=0.6246;P<0.001)。

The molecular genetic evaluation of the collected 33 germplasm resources in six different geographical populations were analyzed by SRAP molecular marker,in order to provide reference for the comprehensive utilization of Bletilla striata Rchb.f variety resources,in situ conservation and ex situ conservation of wild resources.The results showed that the genetic diversity of Bletilla striata was abundant at the species level.The genetic diversity index of Bletilla striata populations was Ht=0.3267,Hs=0.2356,Gst=0.2506,Nm=1.4952.These results indicated that there was a mount of gene exchange among the populations,and the genetic variation mainly occurred within the populations.Based on UPGMA,six populations gathered in two clusters.Population structure analysis showed that the best classification result when K=2 was used.Mantel test confirmed that genetic distance of species was significantly positive correlated with geographical distance(r=0.6246;P<0.001).
