NaCl可促进HD2家族基因对拟南芥根长发育的调控

HD2 Gene Family Can Modulate Root Length Development under NaCl Stresses in Arabidopsis

摘要 本论文通过拟南芥HD2家族基因突变体和转基因过表达株系研究了该家族成员对主根发育的影响,以及对NaCl处理的响应。结果显示,在正常培养条件下,hd2q突变体可以导致主根缩短5.29%,而HD2过表达株系可以促进主根伸长12.94%~28.24%。100 mmol/L NaCl处理后,上述表型差异更加显著。此外,在主根的根尖处,HD2家族成员均有蛋白积累,经过100 mmol/L NaCl处理后,4个成员在根中的转录水平和蛋白积累水平都有显著提高,其中HD2A最为明显。研究表明,HD2家族成员在正常培养和盐处理条件下均能促进主根的伸长,这可能与HD2影响分生区细胞的分裂和伸长区细胞的伸长有关。该研究为进一步探究HD2家族基因在植物根系发育和逆境响应中的作用提供了基础。 This study investigated the effects of HD2 gene family members on the primary root development and its responses to the salt treatment using an Arabidopsis thaliana HD2 family gene mutant and transgenic overexpression lines.Under the normal growth condition,the hd2q mutant resulted in a shorter primary root(5.29%reduction),while the HD2 overexpression lines promoted the elongation of the primary roots(12.94%to 28.24%increase).These phenotypic differences were even more pronounced after the treatment with 100 mmol/L NaCl.Additionally,HD2 family members accumulated proteins at the root tips,and their transcription and protein accumulation levels in the roots that significantly increased after NaCl treatment,with HD2A being substantially affected.These results indicate that the HD2 family members can promote the elongation of the primary roots under the normal growth and salt treatment conditions,which may relate to their effects on the cell division or elongation in the meristems and elongation zones.This study provides a basis for further exploring the role of the HD2 family genes in the plant root development and responses to stresses.
