摘要
为揭示油茶(Camellia oleifera)植株内生真菌组织分布特点,筛选具有抗菌和促生活性的内生真菌资源,对油茶3~4年生植株的根、茎、叶等组织器官中的内生真菌进行了分离鉴定,并进行了体外拮抗活性和分泌IAA能力、解磷能力、产嗜铁素能力等生化活性测定。共分离获得内生真菌26株,其中从根中分离的内生真菌占总数的50%,从根和叶中分离的内生真菌优势菌属分别为土赤壳属(Ilyonectria)和叶点霉属(Phyllosticta)。菌株Ro18对灰霉菌(Botrytis cinerea)和果生炭疽病菌(Colletotrichum fructicola)的生长有明显抑制作用。菌株Ro03同时具有分泌IAA、解磷、产嗜铁素等生物活性。菌株iL04和Ro10可以促进拟南芥(Arabidopsis thaliana)幼苗的侧根生长,其侧根数量分别为28和16,多于空白对照组的;菌株iL04、iL05、Ro03可以增加拟南芥幼苗生物量的累积,拟南芥幼苗平均鲜重分别为28.43、23.87、28.95mg,高于对照组的。菌株iL04、Ro03、Ro18作为生物防治与促生功能菌株的应用潜力巨大,发展前景广阔。
In order to reveal the tissue distribution characteristics of endophytic fungi in Camellia oleifera and screen endophytic fungal resources with antibacterial and growth-promoting properties,the endophytic fungi in roots,stems,and leaves of 3-to 4-year-old C.oleifera plants were isolated and identified.The antagonistic activity,IAA secretion ability,phosphorus solubilizing ability and ferrophilin production ability were detemined in vitro.A total of 26 strains of endophytic fungi were isolated,of which 50%were isolated from the roots.The dominant endophytic fungi genera in the roots and leaves were lyonectria and Phylosticta,respectively.Strain Ro18 significantly inhibited the growth of Botrytis cinerea and Colletotrichum fructicola.Strain Ro03 also had biological activities such as IAA secretion,phosphorus solubilization,and siderophile production.Strain iL04 and Ro10 could promote the lateral root growth of Arabidopsis thaliana seedlings,and the number of lateral roots was 28 and 16,respectively,which was more than that of the blank control group.Strain iL04,iL05 and Ro03 could increase the biomass accumulation of A.thaliana seedlings,with average fresh weight of the seedlings was 28.43,23.87,and 28.95 mg,respectively,higher than that of the control group.Strains iL04,Ro03 and Ro18 had great potential and broad development prospects as biocontrol and growth-promoting strains.
