摘要
目的:研究微波加热和传统加热对三七提取物的药效差异。方法:采用超高效液相色谱法分别检测2种加热方式三七提取物中7种成分的含量,以提取物浓度-时间绘制曲线图,分析达到平衡浓度的时间;分别采用腹腔注射盐酸异丙肾上腺素(isoproterenol,ISO)建立大鼠急性心肌缺血模型、腹腔注射ISO和冰水浴建立大鼠急性血瘀模型,通过测定血清、血浆及心肌组织中药效指标因子,观察心肌组织病理变化,运用模糊综合评判法对药效指标进行综合评分,评价2种加热方式药效差异。结果:微波加热20 min时三七中7种成分已达到平衡浓度,而传统加热最佳工艺需要180 min;微波加热20 min与传统加热180 min三七提取物药效学指标没有显著差异,但微波加热提取物药效指标综合评分(76.50分)大于传统加热(51.63分)。结论:微波加热提取三七不仅快速高效,且不影响药效,是一种极具发展潜力值得推广普及的提取新方法。
OBJECTIVE To explore the differences between microwave heating and traditional heating on the pharmacodynamics of Panax notoginseng(PN)extract.METHODS Ultra-performance liquid chromatography was utilized for detecting the contents of 7 components in PN extract by two heating modes.And extract concentration-time curve was plotted for measuring time to reach the equilibrium concentration.Acute myocardial ischemia in rats was induced through an intraperitoneal injection of isoproterenol(ISO).And acute blood stasis was induced through combining ISO injection with an ice water bath.The pharmacodynamic parameters of serum,plasma and myocardial tissues were measured.The pathological changes of myocardial tissues were observed.Fuzzy comprehensive evaluation method was utilized for comprehensively scoring the efficacy indices to evaluate the differences in efficacy between two heating modes.RESULTS The equilibrium concentration of seven saponins was attained at 20 min by microwave heating.However,the optimal process of traditional heating required 180 min.No significant difference in pharmacodynamic index existed between microwave heating and traditional heating.However,the score of pharmacodynamic index by microwave heating was higher than that by traditional heating(76.50 vs.51.63).CONCLUSION Microwave heating extraction of PN extract is both rapid and efficient.And it has no effect upon pharmacodynamics.This novel extraction technique exhibits great potentials for further developments and merits wider popularization.
