陆地棉YTH基因家族的鉴定与表达特征分析

摘要 [目的]YT521-B同源(YTH)结构域蛋白可识别结合N6-甲基腺苷(m6A)调控mRNA的代谢过程,在植物生长发育与逆境响应中发挥重要作用。探究棉花YTH家族基因的组织表达模式、响应非生物胁迫和外源激素的表达特征,可为深入研究棉花YTH基因功能提供理论依据。[方法]采用生物信息学方法对陆地棉YTH基因家族进行全基因组分析,并利用转录组数据和实时荧光定量PCR(qPCR)分析GhYTH基因在逆境胁迫和外源激素处理下的表达特征。[结果]陆地棉中共鉴定获得26个GhYTH基因,在16条染色体上呈现不均一分布,其中22个YTH-DF和4个YTH-DC亚家族成员,GhYTHs具有相似的基序和结构。串联重复和片段复制是驱动陆地棉YTH基因家族扩张的主要方式;陆地棉与拟南芥YTH共线性基因间Ka/Ks<0.5,表明该基因家族在进化过程中受到了较强的纯化选择。转录组结果显示多数GhYTH具有组织表达特异性,GhYTH2/16在各组织中高表达;逆境胁迫可显著增加GhYTH的表达,尤其是GhYTH9/12/14,qPCR结果与转录组结果一致;外源赤霉素、脱落酸和茉莉酸甲酯处理时,GhYTH10/12/14的表达显著增加。[结论]本研究通过陆地棉全基因组分析,获得了5个重要的YTH候选基因GhYTH2/9/10/12/14,它们在棉花生长发育和逆境抵抗中可能发挥着重要的作用,为进一步解析YTH介导的m6A修饰的作用提供了重要的理论依据。 [Objective]The YT521-B homology(YTH)family proteins can recognize and bind N6-methyl-adenosine(m6A),regulating the mRNA metabolism process,playing an important role in plant growth,de-velopment,and stress response.Exploring the tissue expression patterns and response of the cotton YTH gene family,as well as their response to exogenous hormones and abiotic stresses treatments,can provide a theoretical basis for a deeper understanding of the function of cotton GhYTHs genes.[Methods]In this study,the genome-wide identification of YTH genes in Gossypium hirsutum was performed and the basic characteris-tics were analyzed using bioinformatics methods.The expression patterns of GhYTH genes under abiotic stress and exogenous hormone treatment were analyzed using online RNA-Seq data and/or quantitative real-time PCR.[Results]The results showed that a total of 26 GhYTH genes were identified from G.hirsutum,unevenly distributed on 16 chromosomes,including 22 members of the YTH-DF subfamily and four mem⁃bers of the YTH-DC subfamily.GhYTHs exhibited similar motifs and gene structures.Tandem duplication and fragment duplication were the main driving forces for expansion of YTH gene family in G.hirsutum.The Ka/Ks ratio of the collinear genes between G.hirsutum and Arabidopsis were less than 0.5,indicating strong purifying selection during the evolution process of this gene family.Transcriptome results showed most GhYTHs showed tissue-specific expression patterns,with GhYTH2/16 showing high expression in various tissues.Abiotic stress significantly increased the expression of GhYTHs,especially GhYTH9/12/14,which was consistent with the qPCR results.Additionally,exogenous treatments with gibberellin,abscisic acid and methyl jasmonate led to a significant increase in the expression of GhYTH10/12/14.[Conclusion]Through whole-genome analysis of G.hirsutum,five important candidate genes,including GhYTH2/9/10/12/14,were identified.They may play important roles in cotton growth,development,and stress resistance,provid⁃ing important theoretical basis for further research on the role of YTH-mediated m6A modification in cotton.
