NR4A1在食管癌中的表达及其临床预后意义

Expression and Clinical Prognostic Significance of NR4A1 in Esophageal Cancer

目的研究核受体NR4A1在食管癌(esophageal cancer,EC)中表达、临床意义和对EC细胞增殖的影响。方法基于在线数据库分析NR4A1在EC中的表达水平、临床特征相关性、预后影响及免疫细胞浸润情况,通过CCK-8和克隆形成实验探索NR4A1对EC细胞增殖的影响。结果NR4A1在EC组织中高表达且与病理分型、临床分期、肿瘤分级、淋巴结转移情况、差OS显著相关(P<0.05),免疫浸润分析提示EC中NR4A1表达与CD4^(+)T细胞、CD8^(+)T细胞、B细胞、巨噬细胞和自然杀伤细胞的富集相关(P<0.05),且下调NR4A1的表达可显著抑制细胞增殖(P<0.05)。结论NR4A1在EC中表达升高,且高表达患者预后较差,抑制NR4A1表达可阻止EC细胞增殖。

Objective To study the expression of NR4A1 and its clinical significance and effects on the proliferation in EC.Methods The expression level of NR4A1,its correlation of clinical features,prognostic effects and immune cell infiltration in EC were analyzed based on online database analysis,and the impact of NR4A1 on the proliferation of EC cells was explored by CCK-8 and clonal formation experiments.Results NR4A1 was highly expressed in EC and was significantly correlated with pathological classification,clinical stage,tumor grade,lymph node metastasis and short OS(P<0.05),immunoinfiltration analysis suggested that NR4A1 expression was correlated with the enrichment of CD4^(+)T cells,CD8^(+)T cells,B cells,macrophages and natural killer cells(P<0.05),and inhibiting the expression of NR4A1 could significantly inhibit the proliferation of EC cells(P<0.05).Conclusion The expression of NR4A1 is increased in EC and the prognosis of patients with high expression is poor.In addition,inhibition of NR4A1 expression can prevent the proliferation of EC cells.
