摘要
建立了胶囊类保健食品中红景天苷和酪醇含量的测定方法.采用高效液相色谱法(HPLC),以Thermo Scientific AcclaimTM 120 C18(250 mm×4.6 mm, 5μm)为色谱柱,0.01 mol/L乙酸铵溶液-甲醇(V(乙酸铵溶液)∶V(甲醇)=80∶20)为流动相,流速1.0 mL/min,检测波长220 nm.结果表明,红景天苷的色谱峰面积与质量浓度(0.010 43~0.521 5 mg/mL)和酪醇的色谱峰面积与质量浓度(0.009 9~0.198 8 mg/mL)均呈良好线性关系,相关系数(r)分别为0.999 7,0.999 8.红景天苷和酪醇的平均加标回收率分别为100.17%,100.06%,RSD分别为1.05%,1.61%.该方法稳定可靠、切实可行、操作简便、结果准确,可用于胶囊类保健食品中红景天苷和酪醇的含量测定.
A method for the determination of salidroside and tyrosol in capsule health food was established.With Thermo Scientific AcclaimTM 120 C18(250 mm×4.6 mm,5μm)as chromatographic column,0.01 mol/L ammonium acetate solution-methanol(volume ratioof 80:20)as the mobile phase,flow rate of 1.0 mL/min,and detection wavelength of 220 nm,the HPLC results showed that there was a good linear relationship between the peak area and mass concentration(0.01043~0.5215 mg/mL for salidroside and 0.0099~0.1988 mg/mL for ty rosol),with the correlation coefficients(r)of 0.9995 and 0.9997,respectively.The average recovery rates of salidroside and tyrosol were 100.17%and 100.06%,with RSDs of 1.05%and 1.61%,respectively.This method is stable and reliable,practical and simple operation.Importantly,the result is accurate.It can.be used for the determination of salidroside and tyrosol in capsule health food.
