摘要
目的探讨C1GALT1在胃癌组织中的表达及其对胃癌细胞BGC-823生物学行为的影响。方法采用生物信息学、qRT-PCR和Western blot法分析C1GALT1 mRNA和蛋白在胃癌组织和正常胃黏膜组织、胃癌细胞和正常胃黏膜细胞中的表达,以C1GALT1 cDNA序列为靶点设计siRNA瞬时转染BGC-823细胞,采用Transwell实验检测C1GALT1-siRNA对胃癌BGC-823细胞迁移和侵袭能力的影响。运用Western blot法检测BGC-823细胞在转染C1GALT1-siRNA后上皮-间充质转化(epithelial-mesenchymal transition,EMT)相关蛋白的表达。结果C1GALT1在胃癌组织和细胞(BGC-823、SGC-7901、MGC-803)中呈高表达(P<0.05);干扰BGC-823细胞中C1GALT1表达后,BGC-823细胞迁移和侵袭能力均下降(P<0.05),上皮细胞标志物E-cadherin和Claudin-1蛋白表达水平均升高,间充质细胞标志物vimentin和Slug蛋白表达水平均下降(P<0.05)。结论C1GALT1在胃癌组织和细胞中高表达,C1GALT1基因沉默后能抑制胃癌细胞BGC-823的迁移和侵袭能力,其机制可能与EMT相关。
Purpose To investigate the expression of C1GALT1 in gastric cancer and its effect on the biological behavior of BGC-823 in gastric cancer cells.Methods The expression of C1GALT1 mRNA and protein in gastric cancer tissues and normal gastric mucosa,gastric cancer cells and normal gastric mucosal cells was analyzed by bioinformatics,qRT-PCR and Western blot;the transient transfection of siRNA into BGC-823 cells was designed with C1GALT1 cDNA sequence as the target.Transwell assay was used to detect the effect of C1GALT1-siRNA on the migration and invasion ability of BGC-823 cells in gastric cancer.Western blot method detected the expression of epithelial-mesenchymal transition(EMT)-related proteins in BGC-823 after transfection of C1GALT1-siRNA.Results C1GALT1 was highly expressed in gastric cancer tissues and cell lines BGC-823,SGC-7901 and MGC-803,and the expression levels were positively correlated with gastric cancer pathological stagesⅠandⅡ(P<0.05).After interfering with C1GALT1 in BGC-823 cells,the ability of migration and invasion decreased(P<0.05),epithelial cell markers E-cadherin and Claudin-1 protein expression increased,while mesenchymal cell markers vimentin and Slug protein expression decreased(P<0.05).Conclusion C1GALT1 is highly expressed in gastric cancer tissues and cells,silencing of C1GALT1 can inhibit migration and invasion ability of gastric cancer,the mechanism may be related to EMT.
