超声波辅助提取沙棘有机酸及其成分分析

Ultrasound-Assisted Extraction of Organic Acids from Sea Buckthorn and Analysis of Their Components

摘要 通过超声波辅助提取沙棘有机酸,探究超声时间、料液比、超声功率及乙醇体积分数对提取沙棘有机酸得率影响,用响应面优化得到最佳提取条件;采用高效液相色谱法测定超声波辅助提取液中有机酸种类及含量的差异。结果表明:超声波辅助提取的最佳条件为超声时间30 min、料液比1∶15 (g/mL)、超声功率640 W、乙醇体积分数75%,此条件下沙棘总有机酸得率为37.66 mg/g;采用高效液相色谱(high-performance liquid chromatography,HPLC)测定提取液中主要有机酸含量,共检测出6种有机酸,分别是草酸、酒石酸、苹果酸、抗坏血酸、乳酸、柠檬酸,其中酒石酸含量最高(14.45 mg/g),其次为苹果酸(13.11 mg/g)。该研究为沙棘资源高效利用及相关产品开发提供了理论基础。 The organic acids in sea buckthorn were extracted by ultrasonic assisted extraction. Explore the effects of ultrasonic time, liquid/feed ratio, ultrasonic power and volume fraction of ethanol on the yield of the total organic acids extracted. The optimal extraction conditions were obtained by response surface optimization. A comparison of the organic acids' types and contents in ultrasonic-assisted extracts was conducted. The results showed that the perfect circumstances for ultrasoundassisted extraction were 30 minutes of ultrasound, a liquid/feed ratio of 1∶15(g/mL), 640 W of ultrasound power and 75%ethanol volume fraction under which a total organic acid yield was 37.66 mg/g. The main organic acids in the extract were determined by high-performance liquid chromatography(HPLC), and a total of 6 oxalic acids were extracted, namely oxalic acid, tartaric acid, malic acid, ascorbic acid, lactic acid and citric acid being detected. Of these, tartaric acid had the greatest concentration(14.45 mg/g), followed by malic acid(13.11 mg/g). This study provided a theoretical basis for the efficient utilization of sea buckthorn resources and the development of related products.
