ZWINT表达水平对前列腺癌细胞增殖、迁移和侵袭的影响

Effects of the expression level of ZWINT on proliferation,migration,and invasion of prostate cancer cells

摘要 目的探讨着丝粒相关蛋白(ZWINT)表达水平对前列腺癌细胞增殖、迁移及侵袭的影响。方法通过生物信息学分析ZWINT在前列腺癌组织中的表达水平以及与预后相关性,并进一步在正常前列腺细胞(RWPE⁃1)以及前列腺癌细胞系(LNcap、PC3及DU145)中验证其表达差异性。在LNCap及PC3细胞中分别构建ZWINT敲减组(sh⁃ZWINT),使用CCK⁃8测定sh⁃ZWINT组与阴性对照组之间的细胞增殖能力。伤口愈合实验以及Transwell实验用于分析各组细胞迁移能力。结果相比于癌旁组织及RWPE⁃1,ZWINT在前列腺癌细胞系中过表达。在LNcap及PC3细胞中,敲减ZWINT可以显著减少其增殖活力以及迁移侵袭数量(P<0.05)。结论ZWINT在前列腺癌组织中显著上调,敲除ZWINT可以显著抑制前列腺癌细胞的增殖、迁移侵袭,有望成为前列腺癌预后的潜在生物标志物。 Objective To explore the effects of the expression level of mitogen⁃associated protein(ZWINT)on the proliferation,migration,and invasion abilities of prostate cancer cells.Methods The expression level of ZWINT in prostate cancer tissues and its prognosis value were evaluated using TCGA database and bioinformatics methods,and its differential expression was further confirmed between normal prostate cells(RWPE⁃1)and prostate cancer cells(LNcap,PC3,and DU145).LNCap cells with ZWINT downregulation and PC3 cells with ZWINT downregulation were constructed separately,and their cell proliferation ability was measured using CCK⁃8 assay,and their migration and invasion abilities were analyzed by wound healing assay and transwell invasion assay.Results The expression level of ZWINT was significantly upregulated in prostate cancer and prostate cancer cells compared with that in paracancerous tissues and RWPE⁃1 cells.ZWINT downregulation obviously attenuated the proliferation,migration,and invasion abilities of LNcap and PC3 cells,(P<0.05).Conclusion ZWINT highly expressed in prostate cancer tissues,and knockdown of ZWINT significantly inhibit the proliferation,migratory,and invasion abilities of prostate cancer cells,which is expected to be a potential biomarker for prognosis.
