基于性别差异的少棘巨蜈蚣蛋白组和转录组联合分析

The combined analysis of proteome and transcriptome of the Scolopendra subspinipes mutilans based on sexual differences

摘要 目的通过对不同性别蜈蚣个体进行转录和蛋白关联分析,探究了雌雄个体之间的差异,为其可能存在的药效差异及遗传改良提供依据。方法以12条蜈蚣(雌雄各6条)作为实验材料,基于高通量测序平台Illumina NovaSeq转录组测序技术和蛋白组定量分析技术在转录组和蛋白组水平进行分析,根据差异表达基因(differential expression genes,DEGs)和差异表达蛋白(differential abundance proteins,DAPs)结果对其进行关联分析。结果在蜈蚣雌雄组间共得到3602个DEGs,在雄性中2898个下调,704个上调。雌雄组间蛋白组表达差异分析显示,共有354个DAPs,在雄性中81个上调,273个下调。在对蜈蚣转录组蛋白组的单独分析及联合中发现,雌雄个体在毒素活性、新陈代谢、胆固醇代谢、脂肪酸代谢及合成等相关的基因均呈现明显差异。结论蜈蚣雌雄个体差异表达主要集中在与能量储备、营养物质代谢以及毒素活性等相关通路中,这些差异可能会影响雌雄个体的生长速度及药材品质。 Objective Through transcriptomic and proteomic association analysis of individual males and females of the Scoropendra subspinipes mutilans L.Koch,this article explored the differences between male and female individuals,providing a basis for possible differences in pharmacological effects and genetic improvement.Methods 12 S.mutilans(6 males and 6 females)were used as the experimental material,we conducted transcriptomic sequencing based on the high-throughput sequencing platform Illumina NovaSeq and quantitative analysis of the proteome using Data Independent Acquisition(DIA)technology.Differential expression genes(DEGs)and differential abundance proteins(DAPs)were subjected to correlation analysis.Results A total of 3602 significantly DEGs were identified between male and female individuals of the S.mutilans.Among these,2898 genes were down-regulated and 704 genes up-regulated were in males.Analysis of differential protein abundance between male and female individuals revealed 354 DAPs,with 81 up-regulated and 273 down-regulated proteins in males.In separate and combined analyses of the transcriptome and proteome of the S.mutilans,it was found that male and female individuals exhibited significant differences in genes related to toxin activity,metabolism,cholesterol metabolism,fatty acid metabolism,and synthesis.Conclusion The differential expression between male and female individuals of the S.mutilans mainly focuses on pathways related to energy storage,nutrient metabolism,and toxin activity,and these differences may impact the growth rates and medicinal quality of male and female individuals.
