摘要
目的 探讨HCMV感染患儿外周血中TLR9 mRNA的表达。方法 选择2019年1月-2019年12月江西省儿童医院收治HCMV感染88例为研究对象,其中未给予更昔洛韦治疗23例为治疗前组,给予更昔洛韦治疗27例为治疗后组;同期未感染HCMV健康儿童38例为正常组。采用RT-PCR检测TLR9 mRNA表达,以2^(-△△Ct)方法对mRNA表达进行相对定量,对比分析各组指标的变化。结果 HCMV感染组和正常组外周血中TLR9 mRNA表达量平均值分别为1.986±0.306和3.001±0.543,2组间差异无统计学意义(t=1.724,P=0.088)。HCMV感染治疗前组和治疗后组外周血中TLR9 mRNA表达量平均值分别为1.778±0.312和2.163±0.504,2组间差异无统计学意义(t=0.624,P=0.536)。结论 TLR9 mRNA在HCMV感染儿童外周血中的表达降低,但其表达水平与不同治疗时期无相关性。
Objective To explore the expression of TLR9 mRNA in peripheral blood of children with HCMV infection.Methods 88 children with HCMV infection who were admitted to Jiangxi Provincial Children s Hospital from January 2019 to December 2019 were selected as the study subjects.Among them,23 cases did not received ganciclovir treatment in the pre-treatment group and 27 cases received ganciclovir treatment in the post-treatment group.38 healthy children without HCMV infection in the same period were in normal group.RT-PCR was used to detect the expression of TLR9 mRNA,2^(-ΔΔCt )method was used to quantify the mRNA expression.The changes of indicators in each group were compared and analyzed.Results The average value of TLR9 mRNA expression in peripheral blood of patients in HCMV infection group and normal group was 1.986±0.306 and 3.001±0.543,respectively,with no statistical significance between the two groups(t=1.724,P=0.088).The average value of TLR9 mRNA expression level in peripheral blood of patients with HCMV infection in pre-treatment group and post-treatment group was 1.778±0.312 and 2.163±0.504,respectively,with no statistical significance(t=0.624,P=0.536).Conclusions The expression of TLR9 mRNA in peripheral blood cells of HCMV-infected children is decreased,but its expression levels of TLR9 mRNA has not correlation with different treatment periods.
