新疆库车市和皮山县驴乳源金黄色葡萄球菌的MLST分型、毒力基因和耐药性检测

MLST typing,virulence genes and drug resistance detection of donkey milk-derived Staphylococcus aureus in Kuga City and Pishan County,Xinjiang uyghur autonomous region

摘要 为了了解驴乳源金黄色葡萄球菌(Staphylococcus aureus)的流行情况,试验对从新疆驴乳中分离得到的金黄色葡萄球菌进行多位点序列分型(MLST分型)、药敏试验,以及生物被膜形成能力、毒力基因和溶血活性检测。结果表明:金黄色葡萄球菌在健康驴乳中的分离率为8%(6/75),6株金黄色葡萄球菌分别属于5个MLST分型(STs),即ST6238、ST8115、ST8116、ST8126和ST1660(其中ST8115、ST8116和ST8126为新发现的STs)。所有分离株对青霉素、克林霉素、红霉素和利福平等有不同程度的耐受,其中4株为耐甲氧西林的金黄色葡萄球菌(MRSA),并且有2株MRSA对利奈唑胺耐药;4株金黄色葡萄球菌具有生物被膜形成能力。在驴乳金黄色葡萄球菌中检测到11种毒力基因,分别为sea、sec、seh、sei、tsst-1、hla、hlb、eta、clfA、clfB和cna基因,并且均具有溶血活性。说明驴乳源金黄色葡萄球菌具有较强的耐药性,而且在宿主致病和食品生产安全等方面具有潜在威胁。 In order to understand the prevalence of donkey milk-derived Staphylococcus aureus(S.aureus),in the experiment,donkey milk-derived S.aureus in Xinjiang Uyghur Autonomous Region was subjected to multilocus sequence typing(MLST typing),drug sensitivity test,and detection of biofilm forming ability,virulence genes and hemolytic activity.The results showed that the isolation rate of S.aureus in healthy raw donkey milk was 8%(6/75),and the six S.aureus strains belonged to five STs,namely,ST6238,ST8115,ST8116,ST8126,and ST1660(among which,ST8115,ST8116,and ST8126 were new STs).All isolates were resistant to penicillin,clindamycin,erythromycin and rifampicin in various degrees,four of which were MRSA strains and two MRSA strains were resistant to linezolid,and four S.aureus strains were biofilm-forming.Eleven virulence genes were detected in donkey milk-derived S.aureus,namely sea,sec,seh,sei,tsst-1,hla,hlb,eta,clfA,clfB and cna genes,and all of them had hemolytic activity.The results indicated that donkey milk-derived S.aureus was highly resistant and was a potential threat in terms of host pathogenicity and food production safety.
