摘要
目的探讨聚焦细胞株检测对骨肉瘤细胞侵袭、转移的影响。方法检测Twist1(Twist1基因是一种编码转录因子的基因,其异常活化与多种癌症的发生和发展密切相关。)在143B、MG63、TE85三种不同骨肉瘤中的基础表达,采用划痕实验检测细胞的平行迁移能力,Transwell实验检测细胞的垂直迁移及侵袭能力。结果不同恶性程度的骨肉瘤细胞其划痕愈合率也不同,其中143B细胞最高,MG63细胞次之,TE85细胞最低。通过Twist1表达的诱导能够提高细胞的划痕愈合率,促进细胞的迁移,而抑制Twist1表达则会影响到细胞划痕愈合率,限制细胞迁移;不同细胞恶性程度不同,细胞的侵袭能力也存在一定差别,其中具有最强侵袭能力的细胞是143B,其次是TE85细胞,最低的是MG63细胞。骨肉瘤是一种高度恶性的肿瘤,其恶化程度与Twist1的表达水平密切相关。研究显示,通过诱导Twist1的过表达,可以显著提高骨肉瘤细胞的侵袭能力,尤其是MG63细胞,其侵袭能力的提升尤为明显。这一发现揭示了Twist1在骨肉瘤发展中的关键作用,进一步证实了Twist1的表达水平与骨肉瘤的恶性程度正相关。此外,Twist1的过表达还能够增强骨肉瘤细胞的迁移能力,这对于骨肉瘤的扩散和转移具有重要意义。因此,Twist1可能成为骨肉瘤治疗的一个新的靶点,通过抑制Twist1的表达或功能,可能有助于抑制骨肉瘤的生长和扩散。
Objective To investigate the relationship of focused cell line assays on osteosarcoma cell invasion and metastasis.Methods The basic expression of Twist1 in three different kinds of osteosarcoma(143B,MG63,TE85) was detected.The parallel migration ability of Twist1 cells was detected by scratch test,and the vertical migration and invasion ability of Twist1 cells were detected by Transwell test.Results The basal expression of Twist1 in osteosarcoma cells was significantly higher than that in C3H10 cells,and the difference was statistically significant,the cell with the strongest invasive ability was 143B,followed by TE85 cells and the lowest was MG63 cells.Osteosarcoma is a highly malignant tumor,and its degree of deterioration is closely related to the expression level of Twist1.Studies have shown that the invasive ability of osteosarcoma cells can be significantly enhanced by inducing the overexpression of Twist1,especially in MG63 cells.This finding reveals the critical role of Twist1 in the development of osteosarcoma and further confirms that the expression level of Twist1 is positively correlated with the malignancy of osteosarcoma.In addition,overexpression of Twist1 was able to enhance the migration ability of osteosarcoma cells,which is important for the spread and metastasis of osteosarcoma.Therefore,Twist1 may become a new target for osteosarcoma therapy,and by inhibiting the expression or function of Twist1,it may help to inhibit the growth and spread of osteosarcoma.
