摘要
为了解我国野生水禽中禽副黏病毒6型(APMV-6)的生物学特性,对2020年分离的1株野鸭源APMV-6(Wild duck/Shandong/W683/2020)进行了基因组测序及序列分析。结果显示:该APMV-6分离株基因组为单股负链RNA,基因组长度为16230 nt,基因组结构为3'-N-P-M-F-SH-HN-L-5',3'前导序列和5'尾随序列长度分别为55、54 nt,各基因间隔序列长度不等(2~63 nt);与APMV-1疫苗株La Sota的R值为0.167,抗原性存在较大差异;F蛋白裂解位点为^(104)IREPR↓L^(109),具有典型低致病性APMV分子特征。同源性分析发现,与台湾分离株(MZ802803)同源性最高(96.0%),与其他血清型APMV同源性仅为63.1%~70.2%,与我国雁形目APMV-6分离株(MZ802803)位于同一分支。本研究为进一步了解我国野鸟源APMV-6基因组和生物学特性奠定了基础。
In order to identify the biological characteristics of avian paramyxovirus type 6(APMV-6)present in wild waterfowl in China,one APMV-6 strain(wild duck/Shandong/W683/2020)isolated in 2020 was sequenced and analyzed.The results showed that the genome of the isolate was a single negative-strand RNA with the genome length of 16230 nt and the structure of 3'-N-P-M-F-SH-HN-L-5',the length of 3'leader sequence and 5'trailing sequence was 55 and 54 nt,respectively,and the length of each gene interval sequence varied from 2 to 63 nt;the R value between it with vaccine strain La Sota was 0.167,and the antigenicity was greatly different;the cleavage site of F protein was ^(104)IREPR↓L^(109),showing the typical low pathogenic APMV molecular characteristic.Homology analysis showed that it was the highest identical(96.0%)to Taiwan strain(MZ802803),but 63.1%~70.2%to other serotypes of APMV,locating in the same branch as the APMV-6 strain isolated from antheriformes in China(MZ802803).A basis was laid for further understanding of the genome and biological characteristics of APMV-6 derived from wild birds in China.
