摘要
目的:探讨采用微波辅助法提取的紫薯花青素对小鼠的免疫调节作用。方法:选取新鲜紫薯5 g,微波辅助法获得紫薯花青素的粗提物(3.8±0.6)mg。取64只4周龄昆明小鼠为实验对象,分为对照组(生理盐水100 mg·kg^(-1))和不同剂量紫薯花青素低、中、高处理组(40 mg·kg^(-1)、80 mg·kg^(-1)、160 mg·kg^(-1)),每组各16只,每日灌胃处理,30 d后进行小鼠免疫器官指数、腹腔巨噬细胞的吞噬功能以及脾细胞的增殖能力免疫实验测定。结果:与对照组比较,不同剂量紫薯花青素处理组的脾脏指数均显著提高(P<0.05),而胸腺指数差异无统计学意义(P>0.05);不同剂量紫薯花青素处理组的巨噬细胞吞噬率与吞噬指数均明显增强,中高剂量组增加更加显著(P<0.05);随着紫薯花青素剂量的增加,紫薯花青素处理组的脾细胞增殖能力也明显增强(P<0.05)。结论:紫薯花青素能够增强小鼠腹腔巨噬细胞吞噬鸡红细胞的能力,促进巨噬细胞的吞噬功能,增强非特异性免疫水平,提高小鼠脾淋巴细胞的增殖能力;表明紫薯花青素具有一定的免疫调节功能。
Objective:To study the immunoregulation effect of sweet potato anthocyanins in mice by extracting anthocyanins from sweet potato with microwave-assisted method.Methods:A total of 5g fresh sweet potato was selected and the crude extract of anthocyanins of sweet potato(3.8±0.6)mg was obtained by microwave-assisted method.64 four-week-old Kunming mice were divided into the control group(normal saline 100 mg·kg^(-1))and the anthocyanin-treated groups of sweet potato with different doses(40 mg·kg^(-1),80 mg·kg^(-1),160 mg·kg^(-1)).The immune organ coefficient,the phagocytic function of peritoneal macrophages,and the proliferation ability of spleen cells were measured after 30 d.Results:Compared with the control group,the spleen index of sweet potato anthocyanin-treated groups was significantly increased(P<0.05),but there was no significant difference in thymus index.The phagocytosis rate and phagocytic index of macrophages in the anthocyanin-treated groups were also significantly enhanced,and the effect of the medium and high-dose groups was more significant(P<0.05).The proliferation ability of spleen cells in the anthocyanin-treated groups was significantly increased(P<0.05),and with the increase of anthocyanin dose,the effect was more significant.Conclusion:The anthocyanins from sweet potato can enhance the ability of mouse peritoneal macrophages to phagocytize chicken red blood cells,promote the phagocytic function of macrophages,enhance the level of non-specific immunity,and improve the proliferation ability of spleen lymphocytes in mice.It shows that anthocyanins of sweet potato has a certain immunoregulation effect.
