寻常性银屑病患者皮肤菌群差异研究

摘要 目的:阐明贵州地区银屑病患者皮损样本、正常皮肤样本及健康人群皮肤样本之间皮肤定植微生物的差异。方法:纳入15例寻常性银屑病患者及6例健康志愿者,采集患者皮损样本、正常皮肤样本及志愿者健康皮肤样本微生物,提取DNA并扩增细菌、真核微生物、真菌、古细菌扩增子,采用16S rDNA基因测序,分析微生物组成及丰度。结果:银屑病患者皮损部位微生物多样性较银屑病患者正常皮肤微生物高;组成成分分析发现其组成与正常皮肤表面微生物分离,呈现明显的聚类趋势。与银屑病患者正常皮肤微生物组成比较,银屑病患者皮损部位特异性高分布的菌群包括梭状芽胞杆菌纲-梭菌目-瘤胃菌科、梭状芽胞杆菌纲-梭菌目-毛螺旋菌科以及拟杆菌纲-拟杆菌目-拟杆菌科;而微生物低丰度菌群为乳杆菌目-乳酸细菌科以及假单胞菌目-莫拉菌科。结论:相比于银屑病患者正常皮肤定植的微生物,皮损部位微生物呈现特征性改变,提示差异性分布菌群可能是银屑病潜在的生物标志物。 Objective:To elucidate the differences in skin microbial colonization among psoriatic skin lesions,normal skin samples from patients with psoriasis vulgaris,and normal skin samples from healthy individuals in Guizhou province,China.Methods:Sixteen patients with psoriasis vulgaris and 6 healthy volunteers were included in the study.Samples from psoriatic lesion and normal skin of patients with psoriasis,as well as normal skin of healthy volunteers,were collected.Microbial DNA was extracted,followed by amplification of bacterial,eukaryotic,fungal,and archaeal DNA,using 16S rDNA sequencing.Microbial composition and abundance were analyzed.Results:Skin microbiome diversity was higher in lesional skin compared with normal,non-lesional skin in patients with psoriasis.Composition analysis showed distinct clustering of skin microbiota between psoriatic skin lesion and normal skin.Comparison of the microbial composition of psoriatic skin lesion and normal skin of patients with psoriasis revealed the presence of specific bacteria with high distribution in the lesional skin,including Clostridia-Clostridiales-Ruminococcaceae,Clostridia-Clostridiales-Lachnospiraceae and Bacteroidetes-Bacteroidia-bacteroidales.Compared with normal skin,the low-abundance bacteria in psoriatic skin lesions were Lactobacillaceae and Pseudomonadales-Moraxellaceae.Conclusion:Skin microbial colonization in psoriatic skin lesions demonstrates characteristic changes,compared with normal skin,in patients with psoriasis.The differentially distributed microbial communities may serve as potential biomarkers in psoriasis.
