UPLC法测定葛根中9个化学成分的含量

摘要 目的建立葛根的超高效液相色谱法(UPLC)特征图谱及其3'-羟基葛根素、葛根素、3'-甲氧基葛根素、葛根素-6''-O-木糖苷、葛根素芹菜糖苷、大豆苷、染料木苷、芒柄花苷和大豆苷元9个成分的UPLC含量测定方法。方法采用UPLC,Agilent Eclipse Plus C18(RRHD)色谱柱(100 mm×2.1 mm,1.8μm),以0.05%磷酸水溶液-乙腈为流动相,梯度洗脱,流速为0.3 mL/min,检测波长为254 nm。结果建立了葛根的UPLC指纹图谱,并指认出其中9个共有峰,各批次样本相似度均大于0.90,9个成分分别在各自范围内线性关系良好(r>0.9990),平均回收率为103.5%~98.1%,RSD为0.58%~3.19%。结论建立的方法简便、准确、可靠,可以用于葛根的质量评价。 Objective e To establish a method for characteristic chromatogram of Pueraria lobata and to determine the 9 contents of 3'-hydroxypuerarin,puerarin,3'-methoxypuerarin,puerarin 6"-O-xyloside,mirificin,daidzin,genistin,ononin,daidzein using UPLC.Methods UPLC analysis was performed on an Agilent Eclipse Plus Cis(RRHD)column(100 mm×2.1 mm,1.8μm),employing a the gradient elution method with a mobile phase consisting of 0.05%phosphoric acid solution and acetonitrile.The flow rate was set at 0.3 mL/min,and the detection wavelengths was 254 nm.Results UPLC fingerprint spectrum of Pueraria lobata was established and 9 components were identified.The similarities of all batches of samples were all above 0.90 comparing with the reference.9 components had good linear relationships with their respective ranges(r>0.9990),and the average recoveries were 103.5%~98.1%with RSD of 0.58%~3.19%.Conclusion The method of UPLC characteristic chromatograms and the content determination of Pueraria lobata is simple,accurate,reliable,and can be used for its quality control.
