摘要
目的:建立红线茄高效、稳定离体再生体系,为后期遗传转化、基因功能验证及离体诱变育种等提供材料和技术。方法:将红线茄品种‘皖茄15’无菌发芽的种苗的子叶、真叶、下胚轴和茎段4种外植体接入15种激素组合的MS培养基中,筛选合适外植体,继而研究真叶愈伤组织诱导、再分化、生根等环节的激素配比。结果:真叶为合适外植体,6-BA诱导外植体分化能力弱,IAA容易诱导根的形成,而NAA诱导愈伤组织效果较好;真叶愈伤组织诱导最优培养基为MS+0.1 mg/L TDZ+0.2 mg/L NAA,诱导率为100%,生长状态好;愈伤再分化最优培养基为MS+0.1 mg/L TDZ,芽分化率为100%,平均每块愈伤组织产生5.30个健壮芽;芽苗在不加激素MS培养基上生根率达100%,平均根数为4.60;再生植株经过驯化、移栽,成活率达90%以上。结论:本研究以真叶为外植体,经过愈伤组织诱导、再分化、生根、驯化、移栽等过程,建立了红线茄品种‘皖茄15’高效稳定的再生体系。
Objective:To establish an efficient and stable regeneration in vitro of red long eggplant(Solanum melongena L.)to provide materials and technology for genetic transformation,gene function verification,and mutation breeding in vitro.Methods:Cotyledons,euphyllas,hypocotyls,and stem from aseptic sprouting seedlings of variety Wanqie 15 were inoculated into MS containing 15 hormone combination for screening suitable explants.Subsequently,the hormone ratio in euphyllas callus induction,redifferentiation and shoots rooting were studied.Results:Euphyllas was the suitable explant,and the inducing differentiation activity of 6-BA was very weak,and it was easy for IAA to induce rooting,while NAA has a better effect on callus induction.MS+0.1 mg/L T DZ+0.2 mg/L NAA was suitable for euphyllas callus induction,and the induction rate was 100%,the callus grew well.The best medium for callus redifferentiation was MS+0.1 mg/L TDZ,and adventitious bud differentiation rate was 100%,average number of strong differentiation buds was 5.30.The optimum rooting medium was MS with rooting rate reaching 100%,and the average number of roots was 4.60.After domestication and transplantation,the survival rate of regeneration seedling was over 90%.Conclusion:This study established an efficient and stable regeneration system of red long eggplants variety Wanqie 15 by using euphyllas as explants via callus induction,redifferentiation,rooting,domestication,and transplantation.
