基于AHP-EWM和正交设计优选黄芩黄酮提取工艺及抗炎活性分析

Optimized Extraction Process and Anti-inflammatory Activity Analysis of Scutellaria baicalensis Flavonoids Based on AHP-EWM and Orthogonal Design

摘要 目的:优化黄芩(Scutellaria baicalensis Georgi)中黄酮类成分的提取工艺,分析其体外抗炎活性。方法:以乙醇浓度、超声时间、超声温度及料液比为考察因素,以黄芩苷、汉黄芩苷、黄芩素及汉黄芩素为评价指标,在正交试验基础上分别采用层次分析法(AHP)、熵权法(EWM)及层次分析法-熵权法(AHP-EWM)法确定各指标权重系数,优化黄芩中黄酮类成分的提取工艺,并以最佳工艺制备黄芩提取物,利用MTT法和酶联免疫法(ELISA)分析其体外抗炎活性。结果:AHP-EWM法确定的权重系数科学合理,优选的最佳提取条件为:乙醇浓度55%、超声时间25 min、超声温度60℃、料液比1∶25。ELISA法结果表明,黄芩提取物浓度为50~300μg/mL时,可显著抑制RAW264.7炎性细胞TNF-α、IL-6的释放。结论:AHP-EWM法确定的权重系数客观真实,优选的工艺稳定可靠,该工艺下制备的黄芩提取物具有良好的体外抗炎活性。 Objective:To optimize the extraction process of flavonoids from Scutellaria baicalensis Georgi and analyze its anti-inflammatory activity in vitro.Methods:Ethanol concentration,ultrasonic time,ultrasonic temperature and solid-liquid ratio were used as factors.Baicalin,wogonoside baicalein and wogonin were used as evaluation indexes.On the basis of orthogonal experiment,analytic hierarchy process(AHP),entropy weight method(EWM)and analytic hierarchy process-entropy weight method(AHP-EWM)were used to determine the weight coefficients of each index and optimize the extraction process of flavonoids from S.baicalensis.The extract of S.baicalensis was prepared by the optimum technology,and its anti-inflammatory activity in vitro was analyzed by MTT and enzymelinked immunoassay(ELISA).Results:The weight coefficients determined by AHP-EWM method were scientific and reasonable.The optimal extraction conditions were as follows:ethanol concentration of 55%,ultrasonic time of 25 min,ultrasonic temperature of 60℃,solid-liquid ratio of 1∶25.The results of ELISA showed that when the concentration of S.baicalensis extract was 50~300μg/mL,it could significantly inhibit the release of TNF-αand IL-6 in RAW264.7 inflammatory cells.Conclusion:The weight coefficients determined by AHP-EWM method were objective and true.The optimal process were stable and reliable.The extract of S.baicalensis prepared by this technique had good antiinflammatory activity in vitro.
