摘要
目的研究水蛭素对脓毒症大鼠血小板功能的影响。方法将45只大鼠分为假手术组、模型组和水蛭素组。术后6 h和24 h对三组大鼠进行采血并检测血小板计数、血小板聚集率和P-选择素表达率水平。结果水蛭素组术后6 h血小板计数较模型组下降(P<0.05)。模型组术后24 h血小板计数较水蛭素组和假手术组降低(P<0.05),但假手术组术后24 h血小板计数较水蛭素组明显上升(P<0.05)。模型组术后6 h血小板聚集率较假手术组和水蛭素组升高(P<0.05),而水蛭素组术后6 h血小板聚集率较假手术组明显更高(P<0.05)。假手术组术后24 h血小板聚集率较模型组及水蛭素组明显更高(P<0.05)。水蛭素组术后24 h血小板聚集率较模型组更高(P<0.05)。术后6 h及24 h,模型组P-选择素表达率较假手术组及水蛭素组明显增加(P<0.05)。水蛭素组术后6 h及24 h的P-选择素表达率较假手术组更高(P<0.05)。结论水蛭素能提高脓毒症大鼠的血小板数量,增强聚集功能并下调P-选择素表达水平以发挥抗炎作用。
Objective To investigate the effect of hirudin on platelet function in rat with sepsis.Methods A total of 45 healthy rat were randomly divided into a sham operation(SO)group,a model(ML)group,and an hirudin(HI)group.Platelet count,platelet aggregation and P-selectin expression rate were tested at 6 hours and 24 hours after operation.Results The platelet count in HI group was lower than that in ML group at 6 hours after operation(P<0.05).The platelet count in ML group was lower than that in HI group and SO group at 24 hours after operation(P<0.05),but the platelet count in SO group was significantly higher than that in HI group at 24 hours after operation(P<0.05).The platelet aggregation rate in ML group was higher than that in SO group and HI group at 6 hours after op⁃eration(P<0.05),while the platelet aggregation rate in HI group was significantly higher than that in SO group at 6 hours after opera⁃tion(P<0.05).The platelet aggregation rate in SO group was significantly higher than that in ML group and HI group at 24 hours after operation(P<0.05).The platelet aggregation rate in HI group was higher than that in ML group at 24 hours after operation(P<0.05).At 6 hours and 24 hours after operation,the expression of P-selectin in ML group was significantly higher than that in SO group and HI group(P<0.05).The expression rate of P-selectin in HI group at 6 hours and 24 hours after operation was higher than that in SO group(P<0.05).Conclusion Hirudin can increase platelet count,improve aggregation ability and decrease the expression of P-selectin,which finally exert anti-inflammatory effect.
