栅藻多糖的复合酶法提取工艺、化学组成及其抗氧化活性研究

Study on Extraction Process of Complex Enzyme Method,Chemical Composition and Antioxidant Activity of Polysaccharide from Tetranephris brasiliensis

摘要 以栅藻粉为研究对象,研究栅藻多糖(TBP)的复合酶法提取工艺、化学组成及抗氧化活性。采用复合酶法提取TBP,考察酶解pH、复合酶(纤维素酶和木瓜蛋白酶按1∶1复配)添加量、酶解温度、酶解时间对TBP提取率的影响,并通过响应面模型优化TBP的提取工艺。对TBP的化学成分含量、单糖组成、官能团结构和体外抗氧化活性进行了分析研究。实验结果表明,TBP的复合酶法最佳提取工艺参数为酶解pH 6.1、复合酶添加量5.35%、酶解温度53.1℃和酶解时间84.2 min,在最佳提取条件下,TBP提取率可达(8.86±0.13)%;经化学组成分析,发现TBP的总糖含量为(57.59±1.06)%、蛋白质含量为(5.14±0.88)%、糖醛酸含量为(3.16±0.42)%,TBP具备典型的糖类化合物特征峰,含有吡喃糖和硫酸基团,其单糖共计10种类型,主要由葡萄糖、半乳糖、甘露糖组成;对TBP的抗氧化活性进行研究,发现TBP对ABTS自由基、超氧阴离子自由基和羟自由基具有较好的清除能力。该研究可对栅藻多糖的提取及其在食品工业中的应用提供相应参考。 With Tetranephris brasiliensis powder as the research object,the extraction process of complex enzyme method,chemical composition and antioxidant activity of Tetranephris brasiliensis polysaccharide(TBP)are studied.TBP is extracted by complex enzyme method.The effects of enzymatic hydrolysis pH,the addition amount of complex enzymes(the ratio of cellulase to papain of 1∶1),enzymatic hydrolysis temperature and enzymatic hydrolysis time on the extraction rate of TBP are investigated,and response surface model is used to optimize the extraction process of TBP.The content of chemical components,monosaccharide composition,functional group structure and in vitro antioxidant activity of TBP are analyzed and studied.The results show that the optimal extraction process parameters of TBP by complex enzyme method are enzymatic hydrolysis pH of 6.1,addition amount of complex enzymes of 5.35%,enzymatic hydrolysis temperature of 53.1℃and enzymatic hydrolysis time of 84.2 min.The extraction rate of TBP can reach(8.86±0.13)%under the optimal extraction conditions.Through chemical composition analysis,it is found that the total sugar content of TBP is(57.59±1.06)%,protein content is(5.14±0.88)%,and glucuronic acid content is(3.16±0.42)%.TBP has typical characteristic peaks of carbohydrates,and it contains pyranose and sulfate groups,with a total of ten types of monosaccharides,mainly including glucose,galactose and mannose.The antioxidant activity of TBP is studied,and it is found that TBP has good scavenging capacity on ABTS radicals,superoxide anion radicals and hydroxyl radicals.This study can provide references for the extraction of Tetranephris brasiliensis polysaccharide and its application in food industry.
