泌尿生殖道感染沙眼衣原体PmpH基因多态性分析

摘要 目的探讨泌尿生殖道感染沙眼衣原体多态膜蛋白H(PmpH)基因的多态性及与临床感染的关系,为研究PmpH作为候选疫苗在不同人群的应用差异提供理论基础。方法收集泌尿生殖道标本,标本处理后筛选阳性标本,巢式PCR扩增PmpH基因并进行序列测定,比对分析确定多态性位点,构建系统发育树进行基因分型,用卡方检验探讨PmpH基因型与患者性别、年龄的相关性。结果共获得阳性标本86份,70份标本扩增得到PmpH基因。多态性分析共发现21个多态性位点,其中13个位点能引起氨基酸差异。根据系统发育树的结果,将70例标本分为3个基因型。χ^(2)检验结果表明:PmpH基因型与性别没有相关性,与年龄具有相关性,20岁以下患者、30~40岁患者和40岁以上患者中,基因型1型最多,占比57.14%~77.27%;20~30岁患者中,基因型2最多,占比为58.33%。结论本组患者中引起泌尿生殖道感染的沙眼衣原体的PmpH基因共发现21个多态性位点,13个位点能引起氨基酸差异,PmpH基因可以分成3个基因型,基因型与患者年龄具有相关性。 Objective To investigate the polymorphism in the polymorphic membrane protein H(PmpH)gene of Chlamydia trachomatis causing urogenital tract infections,and its relationship with clinical infection,providing a theoretical basis for studying the application of PmpH protein as a vaccine candidate.Methods The urogenital tract specimens were collected and screened for Chlamydia trachomatis presence.The PmpH gene was amplified using nested PCR,sequenced,and analyzed to determine which sites were polymorphic.The phylogenetic tree was constructed,and genotyping was performed.The correlation between PmpH genotype and patient sex and age was determined using a chi-square test.Results A total of 86 positive specimens were obtained,and the PmpH gene was amplified from 70 specimens.A total of 21 polymorphic sites were found,of which 13 could cause amino acid differences.Seventy specimens were divided into three genotypes according to the phylogenetic tree results.The PmpH genotype was correlated with the patient′s age but not sex.Genotype 1 was the most prevalent in patients under 20 years old,patients aged from 30 to 40 years,and patients over 40 years old,accounting for 57.14%to 77.27%of cases.Genotype 2 was the most frequent in patients aged 20 to 30 years,accounting for 58.33%of cases.Conclusion A total of 21 polymorphic sites were found in the PmpH gene of Chlamydia trachomatis in infected patients,of which 13 sites could cause amino acid differences.The PmpH gene was divided into three genotypes,and PmpH genotype was correlated with age of the patient.
