血清连蛋白和闭合蛋白5水平与卒中后抑郁大鼠抑郁样行为的相关性

Correlation between serum levels of Zonulin and Claudin-5 and depressive behavior in rats with post stroke depression

目的:探究卒中后抑郁大鼠血清连蛋白(Zonulin)和闭合蛋白5(Claudin-5)水平与抑郁样行为之间的相关性,为卒中后抑郁的诊疗和疗效评估提供依据。方法:通过MCAO手术结合CUMS的方法,建立卒中后抑郁SD大鼠模型。采用蔗糖水偏好实验和强迫游泳实验测试两组大鼠的快感缺失症状和抑郁症状。采用旷场实验观察两组大鼠的自主运动能力、焦虑和抑郁行为。采用酶联免疫吸附试验(ELISA)测定大鼠血清Zonulin和Claudin-5水平。采用Logistic回归分析血清Zonulin、Claudin-5水平与卒中后抑郁大鼠抑郁样行为的相关性。评价血清Zonulin和Claudin-5水平对卒中后抑郁的早期诊断以及疗效评估价值。结果:相对于对照组,卒中后抑郁组大鼠表现出蔗糖水偏好率显著下降,强迫游泳不同时间显著延长等抑郁样行为,且其血清中Zonulin、Claudin-5水平均显著升高。Logistic回归分析显示,血清中Zonulin、Claudin-5水平与大鼠的蔗糖水偏好率负相关,与大鼠的强迫游泳不动时间正相关。结论:血清Zonulin、Claudin-5水平能够在一定程度上反映大鼠卒中后抑郁样行为严重程度,对卒中后抑郁的早期诊断和和疗效评估可能具有一定的潜在价值。

Objective To explore the correlation between serum Zonulin and Claudin-5 levels and depression-like behavior in post-stroke depressed rats,and to provide evidence for the diagnosis and treatment of post-stroke depression and the evaluation of therapeutic effect.Method The SD rat model of post-stroke depression was established by MCAO combined with CUMS.The symptoms of anhedonia and depression were measured by sucrose water preference test and forced swimming test.Open field test was used to observe the autonomic motor ability,anxiety and depression behavior of the two groups of rats.Serum levels of Zonulin and Claudin-5 were determined by enzyme-linked immunosorbent assay(ELISA).Logistic regression was used to analyze the correlation between serum Zonulin and Claudin-5 levels and depression-like behavior in post-stroke depressed rats.To evaluate the value of serum Zonulin and Claudin-5 levels in early diagnosis and efficacy evaluation of post-stroke depression.Results Compared with the control group,the rats in the post-stroke depression group showed significantly decreased sucrose water preference rate,significantly prolonged forced swimming time and other depression-like behaviors,and the serum levels of Zonulin and Claudin-5 were significantly increased.Logistic regression analysis showed that serum levels of Zonulin and Claudin-5 were negatively correlated with rats′sucrose water preference rate,and positively correlated with rats′forced swimming ignorance of time.Conclusion Serum levels of Zonulin and Claudin-5 can reflect the severity of post-stroke depression-like behavior in rats to a certain extent,and may have potential value for early diagnosis and therapeutic evaluation of post-stroke depression.
