摘要
肿瘤微环境(TME)被认为对包括乳腺癌在内的一系列癌症的进展都有很大影响,癌症相关成纤维细胞(CAFs)是TME中最丰富的成分。研究表明,癌症中的CAFs来源于多种类型的细胞,通过分泌因子、产生外泌体、释放营养物质、重塑细胞外基质和抑制免疫细胞功能,促进恶性肿瘤的产生与进展。CAFs由于其在肿瘤中的特异性分布和独特的生物标志物,具有成为癌症的治疗靶点的潜力,目前临床阻断CAFs对周围细胞作用的药物已经开发出来并在临床试验中应用。进一步了解CAFs的促肿瘤效应与抗肿瘤效应中涉及的机制,可以提供控制某些基质性状为目的的治疗干预的未映射的可能性。进一步操纵CAFs并利用它们的生物学特性还可恢复功能失调的微环境并阻止乳腺癌的进展。
It is believed that tumor microenvironment contribute greatly to the progression of a series of cancers,including breast cancer.Cancer associated fibroblasts(CAFs)are the most abundant components of tumor microenvironment.Research has shown that CAFs in cancer originate from various types of cells,promoting the production and progression of malignant tumors by secreting factors,producing exosomes,releasing nutrients,reshaping the extracellular matrix,and inhibiting immune cell function.Due to their specific distribution in tumors and unique biomarkers,CAFs have the potential to become therapeutic targets for cancer.Currently,drugs that block the action of CAFs on surrounding cells have been developed and applied in clinical trials.Further understanding of the mechanisms involved in the tumor promoting and anti-tumor effects of CAFs can provide the possibility of unmapped therapeutic interventions aimed at controlling some matrix properties,further manipulate CAFs and use their biological characteristics to restore the dysfunctional microenvironment and prevent the progress of breast cancer.
