藏药塞北紫堇对小鼠急性肝损伤的保护作用及机制研究

Study on the Protective Effect and Mechanism of Corydalis impatiens on Carbon Tetrachloride Induced Anti-liver Injury in Mice

摘要 目的:研究塞北紫堇的抗肝损伤作用。方法:采用CCl4诱导小鼠急性肝损伤模型,通过观察肝组织病理切片,测定小鼠肝脏指数,血清中AST、ALT、AKP、LDH、TNF-α、IL-1β指标,以及肝组织中MDA、SOD、GSH指标,考察塞北紫堇提取物、生物碱和非生物碱部位的抗肝损伤活性。结果:除非生物碱部位外的各组均可降低模型小鼠血清中的AST、ALT、AKP、LDH、TNF-α、IL-1β水平和肝组织中MDA含量(P<0.05,P<0.01),升高GSH和SOD水平(P<0.01);肝指数和HE染色结果显示,各组均对肝组织损伤具有保护作用,且效果依次是提取物>生物碱>非生物碱部位。结论:塞北紫堇通过降低脂质过氧化,缓解氧化应激,并抑制炎症因子释放来减少模型小鼠肝损伤,总生物碱为其主要活性部位。 Objective To study the protective effect of Corydalis impatiens on liver injury and its mechanism of action.Methods Carbon tetrachloride was used to induce acute liver injury in mice model.Serum liver function indexes of mice aspertate aminotransferase(AST),alanine aminotransferase(ALT),alkaline phosphatase(AKP),lactate dehydrogenase(LDH),inflammatory factors tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and liver antioxidant stress factors malondialdehyde(MDA),superoxide dismutase(SOD),glutathione(GSH)in each group were detected.The pathological changes of liver were detected by HE staining.Results Compared with model group,SOD and GSH enzyme activities were significantly increased in total extract group and alkaloids group(P<0.01),and the contents of AST,ALT,AKP,LDH,TNF-α,IL-1βand MDA were significantly decreased(P<0.05,P<0.01).The pathological sections showed that Corydalis impatiens could significantly reduced liver damage.Conclusion Corydalis impatiens has a protective effect on CCl 4 induced liver injury,and total alkaloid is the main active part.Its mechanism of action is related to inhibiting inflammatory reation and improving oxidative stress injury of mice liver.
