摘要
为实现多抗生素的便捷、可视化检测,本文以核酸适配体作为分子识别元件,采用ISM技术进行定向进化,利用碳点纳米材料作换能器,采用链置换信号传导技术和表面临近杂交信号的输出策略,研制出了氯霉素和土霉素可视化检测试纸。结果显示,氯霉素和土霉素可视化适配体检测试纸是一种signal-on比色型传感器,检测带的亮度与靶标的浓度正相关。经液质联用法确证,该检测试纸的灵敏度为:土霉素≥24.6μg/L,氯霉素>10.26μg/L。实验结果证明该检测试纸具有经济、快捷、高特异性的优点,有望为政府部门的抗生素风险评估、滥用监管和基层用户的质量控制提供技术支撑。
To realize convenient and visual detection of multiple antibiotics,visual test strip for detection of chloramphenicol and oxytet-racycline is developed using aptamer as molecular recognition element,ISM technology for directed evolution,carbon point nanomaterials as transducers and then integrating signal transmission technology of chain displacement and signal output strategy of surface near the hybridization.Experimental results show that the aptasensor proposed is a signal-on type test paper utilizing colorimetric principle,and the brightness of the test line shows positive correlation with the concentration of the targets.Confirmed by LC-MS/MS,the sensitivity of oxytetracycline is≥24.6μg/L,and that of chloramphenicol is>10.26μg/L.In conclusion,this aptasensor has the advantages of econ-omy,efficiency and high specificity,and is expected to provide technical support for the quality control of primary users and the risk as-sessment and abuse supervision of antibiotics by government.
