橄榄苦苷对LPS诱导小鼠视网膜损伤的保护作用

摘要 为研究橄榄苦苷(oleuropein, OP)对LPS诱导小鼠视网膜损伤的保护作用,将SPFKM小鼠随机分成5组,分别为LPS模型组(LPS)、对照组(Control)、OP高、中、低质量浓度(800、400、200 mg/kg)处理组,连续灌胃21 d,从第14天开始,各组小鼠灌胃30 min后,在异氟烷气体辅助麻醉下,小鼠腹腔注射2.0 mg/kg LPS,对照组小鼠腹腔注射2.0 mg/kg生理盐水,连续7 d,建模完成后,检测小鼠血清中超氧化物歧化酶(superoxide dismutase, SOD)、过氧化氢酶(catalase, CAT)、谷胱甘肽过氧化物酶(glutathione peroxidase, GSH-Px)活性及白细胞介素-1β(IL-1β)和肿瘤坏死因子-α (TNF-α)含量,采用免疫组化SP法检测甘油三脂脂肪酶(PNPLA2)在小鼠视网膜各层结构上的分布特点并观察视网膜组织形态学变化。结果显示,OP使小鼠血清中SOD、CAT和GHS-Px活性显著升高(P<0.01),IL-1β和TNF-α含量下降(P<0.05),视网膜中PNPLA2表达量升高(P<0.05,P<0.01);OP各剂量组均能提高SOD、CAT和GHS-Px活性,降低IL-1β和TNF-α含量,改善LPS对小鼠视网膜的病理损伤,提高视网膜中PNPLA2表达量,具有保护LPS诱导视网膜损伤的作用。 To investigate the protective effect of oleuropein(OP)on LPS-induced retinal damage in mice.The mice were randomly divided into 5 groups:LPS model group(LPS),control group(Control),high,medium,and low dose OP treatment groups(800,400,200 mg/kg),the OP was administered to the mice via intragastric administration for 21 consecutive days,starting from the 14th day.After 30 min of intragastric administration under isoflurane-assisted anesthesia,experimental group mice were intraperitoneally injected with 2.0 mg/kg LPS and control group mice were injected with the same dose of physiological saline for continuous 7 days,after modeling was completed,the activities of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px)and the content of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)in the serum were determined,immunohistochemical SP method was used to study the distribution characteristics of glycerol triglyceride lipase(PNPLA2)in the retina in mice,and observe the morphological changes of retinal tissue.The results showed that OP could significantly improve the activity of SOD,CAT and GSH-Px(P<0.01),reduce the IL-1βand TNF-αcontent(P<0.05)in serum,and increase expression of PNPLA2 in the retina(P<0.05,P<0.01).Oleuropein in all dose groups could improve the activities of SOD,CAT and GSH-Px,reduce the content of IL-1βand TNF-α,improve the pathology of LPS-induced retinal damage,increase the expression of PNPLA2 in the retina.Therefore,it can be concluded that the OP has a protective effect against LPS-induced retinal damage.
