核分裂周期蛋白80复合体通过上皮-间充质转化对乳腺癌细胞侵袭和转移的作用

摘要 目的探究核分裂周期80(NDC80)复合体通过上皮-间充质转化(EMT)对乳腺癌细胞侵袭和转移的作用。方法集取100例乳腺癌的癌旁组织及癌组织,分析癌组织组及癌旁组织组的NDC80、E-cadherin表达,且培养乳腺癌MDA-MB-231细胞,并转染分别为si-NC、si-NDC80、si-E-cadherin;通过RT-PCR法检测mRNA表达,通过Western Blot法对相关蛋白的表达进行测定,通过MTT法、流式细胞仪、划痕实验、Transwell试验对细胞的凋亡、增殖、侵袭及转移予以测定。结果癌组织组的NDC80、E-cadherin表达高于癌旁组织组,差异有统计学意义(P<0.05)。si-NDC80组的NDC80及24、48、72 h细胞增殖率、细胞侵袭数、细胞迁移数低于si-NC组,凋亡率高于si-NC组,差异有统计学意义(P<0.05)。si-E-cadherin组的凋亡率高于si-NC组,细胞迁移数、细胞侵袭数、E-cadherin表达及24、48、72 h细胞增殖率低于si-NC组,差异有统计学意义(P<0.05)。结论NDC80复合体通过调控EMT表达,可有效改善乳腺癌细胞的侵袭、增殖、转移及凋亡,以减缓病情的发展。 Objective To explore the role of the nuclear division cycle 80(NDC 80)complex on breast cancer cell invasion and metastasis through epithelial-mesenchymal transition(EMT).Methods The expression of NDC80 and E-cadherin in 100 cases of breast cancer were analyzed,and MDA-MB-231 cells were cultured and transfected into si-NC,si-NDC80 and si-E-cadherin,respectively.The mRNA expression was measured by RT-PCR,the expression of related proteins was determined by Western Blot,and the apoptosis,proliferation,invasion and metastasis were determined by MTT,flow cytometry,scratch assay,Transwell assay.Results The expressions of NDC80 and E-cadherin in cancer tissue group were higher than those in para-cancer tissue group,and the differences were statistically significant(P<0.05).The rate of NDC80,24,48 and 72 h cell proliferation,cell invasion and cell migration in si-NDC80 group were lower than those in si-NC group,and the apoptosis rate was higher than that in si-NC group,the differences were statistically significant(P<0.05).The apoptosis rate of si-E-cadherin group was higher than that of si-NC group,and the cell migration number,cell invasion number,expression of E-cadherin and cell proliferation rate at 24,48 and 72 h were lower than those of si-NC group,with statistical significances(P<0.05).Conclusion NDC 80 complex can effectively improve the invasion,proliferation,metastasis and apoptosis of breast cancer cells,so as to slow down the development of the disease.
