木犀草素对反复尿路感染大鼠模型尿路上皮屏障的影响

Effect of luteolin on the urinary epithelial barrier in a rat model of recurrent urinary tract infections

摘要 目的探究木犀草素对反复尿路感染大鼠模型尿路上皮屏障的影响.方法选取SPF级SD雌性大鼠45只,其中空白组、模型组、低剂量组、高剂量组各10只;空白组、模型组只进行生理盐水灌胃,不做其他任何处理,低剂量组在模型组的基础上给予木犀草素10 mg/kg灌胃,高剂量组给予40 mg/kg灌胃;检测尿量、尿白细胞水平、炎症因子、分泌型免疫球蛋白A(SIgA)、尿乙酰氨基葡萄糖苷酶(NAG)水平、尿路上皮屏障功能情况及Toll样受体4/核转录因子-κB(TLR4/NF-κB)信号通路相对表达量.结果与空白组比较,模型组、低剂量组、高剂量组尿量情况、E黏钙蛋白(E-cadherin)、闭合小带蛋白-1(ZO-1)水平降低,尿白细胞、白细胞介素-6(IL-6)、IL-8、SIg A、NAG水平、TLR4、NF-κB相对表达量均升高(P<0.05);与模型组相比,低剂量组、高剂量组,尿量、E-cad-herin、ZO-1水平升高,尿白细胞、IL-6、IL-8、SIgA、NAG、TLR4、NF-κB相对表达量均降低(P<0.05);与低剂量组比较,高剂量组尿量、E-cadherin、ZO-1水平升高,尿白细胞、IL-6、IL-8、SIgA、NAG、TLR4、NF-κB相对表达量均降低(P<0.05).结论木犀草素能对反复尿路感染大鼠尿路上皮屏障情况进行改善,减轻炎症情况,提高排尿功能,调节免疫功能,通过调节TLR4/NF-κB信号通路能反映出对反复尿路感染的干预效果. OBJECTIVE To investigate the effects of luteolin on the urothelial epithelial barrier in a rat model of recurrent urinary tract infection.METHODS A total of 45 SPF SD female rats were selected,including blank group,model group,low dose group and high dose group,with 10 rats in each group.The blank group and model group were only given normal saline intragastric administration without any other treatment,while the low-dose group was given 10 mg/kg luteolin intragastric administration on the basis of the model group,and the high-dose group was given 40 mg/kg intragastric administration.Urine volume,urinary white blood cell levels,inflammatory factors,secreted immunoglobulin A(SIgA),urinary acetylglucosaminase(NAG)levels,urinary epithelial barrier function and the relative expression of toll-like receptor 4/nuclear transcription factor-B(TLR4/NF-κB)signaling pathway were measured.RESULTS Compared with the blank group,urine volume,E-cadherin and ZO-1 levels in model group,low dose group and high dose group were decreased,while the levels of leukocyte,interleukin-6 (IL-6), IL-8, SIgA, NAG, TLR4, NF-κB relative expression were increased (P<0.05). Compared with themodel group, urine volume, E-cadherin and ZO-1 levels were increased in the low-dose and high-dose groups, andthe relative expression levels of white blood cells, IL-6, IL-8, SIgA, NAG, TLR4 and NF-κB were decreased(P<0.05). Compared with the low dose group, urine volume, E-cadherin and ZO-1 levels were increased in thehigh dose group, and the levels of white blood cells, IL-6, IL-8, SIgA, NAG, TLR4 and NF-κB relative expressionlevels were decreased (P<0.05). CONCLUSION Luteolin can improve the urothelial epithelial barrier conditionin rats with recurrent urinary tract infection, reduce inflammation, improve voiding function, and regulateimmune function, which can reflect the intervention effect on recurrent urinary tract infection by regulating TLR4/NF-κB signaling pathway.
