摘要
目的探讨中和汤干预脓肿型淋巴结结核的作用机制。方法使用细胞计数试剂盒-8方法检测不同浓度中和汤溶液对人单核细胞(THP-1)的细胞毒性,结果进一步运用二代高通量RNA转录组测序技术进行测序,筛选差异表达基因,并对其进行基因本体论功能注释及京都基因与基因组百科全书通路分析。结果中和汤可降低炎症因子表达,细菌感染相关的通路受抑制,磷脂酰肌醇3-激酶/蛋白激酶B信号通路响应降低,细胞焦亡和自噬相关的信号通路亦受抑制。结论中和汤可能通过抑制磷脂酰肌醇3-激酶/蛋白激酶B信号通路抑制细胞焦亡干预淋巴结结核。
Objective To explore the mechanism of Zhonghe decoction in the intervention of abscess type lymph node tuberculosis.Methods Cell counting Kit 8 method was used to detect the cytotoxicity of THP-1 in different concentrations of neutralization soup solution.The results were further sequenced by second-generation high-throughput RNA transcriptome sequencing technology,and the differentially expressed genes were screened,and the Gene Ontology functional annotation and pathway analysis of Kyoto Encyclopedia of Genes and Genomes were performed.Results Zhonghe decoction can reduce the expression of inflammatory factors,inhibit the pathway related to bacterial infection,reduce the response of phosphatidylinositol 3-kinase/protein kinase B signaling pathway,and inhibit the signaling pathway related to pyroptosis and autophagy.Conclusion Zhonghe decoction may interfere with lymph node tuberculosis by inhibiting cell pyrodeath by inhibiting phosphatidylinositol 3-kinase/protein kinase B signaling pathway.
