摘要
建立同时测定桑叶中新绿原酸、桑皮苷A、绿原酸、隐绿原酸、芦丁、异槲皮苷、烟花苷、异绿原酸C、山奈酚等9种多酚类成分的检测方法。采用高效液相色谱法,使用Shim-pack GIS C_(18)柱(4.6 mm×250 mm,5μm),柱温为30℃,以乙腈-0.2%甲酸水溶液为流动相,进样量10μL,流速为1.0 mL/min^(-1),梯度洗脱,在350 nm波长进行检测。试验结果表明,在85 min内,新绿原酸、桑皮苷A、绿原酸、隐绿原酸、芦丁、异槲皮苷、烟花苷、异绿原酸C、山奈酚能达到良好的分离度,分别在0.0170~0.1068 mg/mL、0.0154~0.1155 mg/mL、0.0146~0.1097 mg/mL、0.0130~0.0977 mg/mL、0.0149~0.1114 mg/mL、0.0136~0.1020 mg/mL、0.0170~0.1273 mg/mL、0.0070~0.0527 mg/mL、0.0151~0.1131 mg/mL、0.0058~0.0435 mg/mL质量浓度范围内呈现良好的线性关系(R为0.9991~0.9998),平均回收率在93.00%~100.51%之间。所建立的方法准确可靠,可用于桑叶的质量控制。
To establish a method forsimultaneous determination of nine polyphenol compounds,including neochlorogenic acid,mulberroside A,chlorogenic acid,cryptochlorogenic acid,rutin,isoquercitrin,nicotiflorin,isochlorogenic acid C and kaempferol.High-performance liquid chromatography equipped with Shim-pack GIS C_(18) column(4.6 mm×250 mm,5μm)was used for polyphenols detection under the condition of 30℃column temperature.Acetonitrile-0.2%formic acid aqueous solution was used as mobile phase.The flow rate is 1.0 ml·min^(-1),10μL extract was injected.Polyphenols was detected at the 350 nm wavelength under the conditions of gradient elution1.Nine polyphenols were absolutely separated within 85 minutes.The linearity ranges of neochlorogenic acid,mulberroside A,chlorogenic acid,cryptochlorogenic acid,rutin,isoquercitrin,nicotiflorin,isochlorogenic acid C and kaempferol were 0.0170~0.1068 mg/mL,0.0154~0.1155 mg/mL,0.0146~0.1097 mg/mL,0.0130~0.0977 mg/mL,0.0149~0.1114 mg/mL,0.0136~0.1020 mg/mL,0.0170~0.1273 mg/mL,0.0070~0.0527 mg/mL,0.0151~0.1131 mg/mL,0.0058~0.0435 mg/mL,respectively.And the r values range from 0.9991 to 0.9998.The average recoveries of nine polyphenols were from 93.00%to 100.51%.the method was proved to be accurate and reliable,and can be used for the quality control of mulberry leaves.
