ABCC4和miR-125b-5p在前列腺癌组织中的表达及意义

Expression and significance of ABCC4 and miR-125b-5p in prostate cancer tissue

摘要 目的检测ABC转运蛋白家族4(ABCC4)、miR-125b-5p在前列腺癌组织中的表达情况,并分析两者与前列腺癌预后的关系。方法选取2019年5月—2021年5月秦皇岛市中医医院收治的经病理组织学诊断确诊的前列腺癌患者60例,前列腺癌新鲜组织标本均在前列腺癌手术中切取,分为前列腺癌组(癌组织)和对照组(癌旁组织)。采用实时荧光定量聚合酶链反应(RT-qPCR)检测前列腺癌组织和癌旁组织中ABCC4 mRNA、miR-125b-5p的表达水平,采用免疫组化法检测组织石蜡切片中ABCC4的表达情况,分析ABCC4、miR-125b-5p表达水平与临床病理特征、前列腺癌患者预后的关系及前列腺癌预后不良的影响因素。计量资料以均数±标准差(x±s)表示,组间比较采用t检验;计数资料组间比较采用χ^(2)检验。采用Pearson法分析前列腺癌组织中ABCC4 mRNA和miR-125b-5p水平的相关性;采用Kaplan-Meier法分析ABCC4、miR-125b-5p表达水平与预后的关系,生存率比较采用Log-rank检验;采用单因素和多因素COX回归分析评价前列腺癌预后不良的影响因素。结果RT-qPCR检测结果显示,前列腺癌组ABCC4 mRNA表达水平高于对照组(P<0.05),miR-125b-5p表达水平低于对照组(P<0.05)。免疫组化法检测结果显示,前列腺癌组织中ABCC4阳性率高于癌旁组织(P<0.05)。Pearson相关性分析结果显示,ABCC4 mRNA表达水平与miR-125b-5p呈负相关(r=-0.687,P<0.05)。前列腺癌患者中ABCC4、miR-125b-5p表达水平与肿瘤分期、血清前列腺特异性抗原、Gleason评分、肿瘤转移有关(P<0.05)。Kaplan-Meier法分析结果显示,ABCC4阳性患者3年累积生存率低于阴性患者(P<0.05),miR-125b-5p高表达患者3年累积生存率高于低表达患者(P<0.05)。多因素COX回归分析结果显示,miR-125b-5p水平偏低、ABCC4水平偏高是前列腺癌不良预后的危险因素(HR=2.685、3.461,P<0.05)。结论ABCC4在前列腺癌患者癌组织中高表达,miR-125b-5p呈低表达,两者与前列腺癌的多种临床病理及预后有关,是前列腺癌患者预后不良的危险因素。 Objective To detect the expression of ABC transporter family 4(ABCC4)and miR-125b-5p in prostate cancer tissues,and analyze their relationship with the prognosis of prostate cancer.Methods From May 2019 to May 2021,60 patients with prostate cancer diagnosed by pathological diagnosis admitted to Qinhuangdao Hospital of Traditional Chinese Medicine were selected.All fresh tissue samples of prostate cancer were obtained from prostate cancer surgery and divided into prostate cancer group(cancer tissue)and control group(adjacent tissue).Real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)was used to detect the expression levels of ABCC4 mRNA and miR-125b-5p in the cancer tissue and adjacent tissue.Immunohistochemistry was used to detect the expression of ABCC4 in paraffin sections of tissues,and to analyze the relationship between the expression levels of ABCC4 and miR-125b-5p and clinicopathological features,prognosis of patients with prostate cancer,and the influencing factors of poor prognosis of prostate cancer.The measurement data were expressed as mean±standard deviation(x±s),and t-test was used for comparison between groups;Chi-square test was used to compare the count data between groups.Pearson method was used to analyze the correlation between ABCC4 mRNA and miR-125b-5p levels in prostate cancer.Kaplan-Meier method was used to analyze the relationship between the expression levels of ABCC4 and miR-125b-5p and prognosis,and Log-rank test was used to compare the survival rates.Univariate and multivariate COX regression analysis was used to evaluate the influencing factors of poor prognosis of prostate cancer.Results The results of RT-qPCR showed that the expression level of ABCC4 mRNA in prostate cancer group was higher than that in control group(P<0.05),and the expression level of miR-125b-5p was lower than that in control group(P<0.05).Immunohistochemistry showed that the positive rate of ABCC4 in prostate cancer tissues was higher than that in adjacent tissues(P<0.05).Pearson correlation analysis showed that the expression level of ABCC4 mRNA was negatively correlated with miR-125b-5p(r=-0.687,P<0.05).The expression level of ABCC4 and miR-125b-5p in patients with prostate cancer was related to tumor stage,serum prostate-specific antigen,Gleason score and tumor metastasis(P<0.05).Kaplan-Meier analysis showed that the 3-year cumulative survival rate of ABCC4-positive patients was lower than that of negative patients(P<0.05),and the 3-year cumulative survival rate of patients with high-expression of miR-125b-5p was higher than that of patients with low-expression(P<0.05).Multivariate COX regression analysis showed that the low level of miR-125b-5p and the high level of ABC-C4 were the risk factors for the poor prognosis of prostate cancer(HR=2.685,3.461,P<0.05).Conclusion The high-expression of ABCC4 and the low-expression of miR-125b-5p in prostate cancer tissues are related to many clinicopathological features and prognosis of prostate cancer,which are risk factors for poor prognosis of prostate cancer patients.
