摘要
为了研究锦鲤sox10b基因及其氨基酸序列的结构与功能,本文以纯红锦鲤皮肤cDNA为模板,使用RT-PCR技术克隆获得了锦鲤sox10b的编码区序列(CDS),并对其氨基酸序列进行了生物信息学分析。结果表明,锦鲤sox10b CDS区全长1452bp,共编码483个氨基酸。氨基酸序列比对显示锦鲤Sox10b具有保守的Sox_N Superfamily结构域和HMG-box_SoxE Superfamily结构域。系统进化树结果显示锦鲤sox10b与鲤sox10b、斑马鱼的sox10亲缘关系最近,锦鲤sox10b可能行使与斑马鱼sox10相似的功能。
In order to explore the structure and function of koi sox10b and its amino acids,the coding sequence(CDS)of koi sox10b was cloned by RT-PCR.Then,the multiple sequences alignment and phylogenetic tree of the amino acid sequences of sox10 among 13 species were conducted.The results showed that the length of koi sox10b CDS was 1452bp,encoding 483 amino acids.Amino acid sequences alignment result showed that koi Sox10b had conserved Sox_N Superfamily domain and HMG-box_SoxE Superfamily domain.The phylogenetic tree showed that koi sox10b was closely related to the common carp sox10b and zebrafish sox10,which suggest the koi sox10b may perform similar functions with zebrafish sox10.
