摘要
目的 建立木瓜饮片高效液相色谱法(HPLC)指纹图谱,对木瓜饮片质量进行较全面的控制。方法 采用HPLC,Kromasil C_(18)色谱柱(250×4.6 mm,5μm),以乙腈-0.4%磷酸溶液为流动相进行梯度洗脱,流速:1.0 ml/min,检测波长:280 nm,柱温:30℃;对13批木瓜饮片HPLC色谱图进行比较分析,评价相似度并进行聚类分析。结果 木瓜饮片的HPLC对照指纹图谱中不同产地样品标定了9个共有峰,除S9外,其他样品相似度均大于0.90;对13份木瓜样品的指纹图谱共有色谱峰的相对峰面积进行系统聚类分析,将13批样品分为3类,分析结果表明,S9与其他样品距离较远。结论 该方法稳定准确,可为提高木瓜饮片整体质量控制提供实验依据。
Objective To establish HPLC fingerprint of Chaenomelis Fructus pieces and control thequality.Methods The method was HPLC.Stationary phase was Kromasoil C_(18) (250×4.6 mm,5μm).Mobile phase consisted of acetonitrile-0.4%H_3PO_(4in) a gradient mode.Flow rate was 1.0 ml/min.Detection wavelength was 210 nm,column temperature was 30℃.The HPLC chromatograms of 13 batches of Chaenomeles were compared and analyzed to evaluate the similarity and cluster analysis.Results Nine common peaks were identified in the HPLC fingerprint of Chaenomeles Fructus pieces.The similarities exceeds 0.90 except sample 9.The relative peak areas of the common chromatographic peaks of 13 papaya samples were analyzed by systematic cluster analysis.The 13 samples were divided into 3 categories.The results showed that S9 was far away from other samples.Conclusion This method was stable,accurate and reproducible,it can be used for the overall quality control of Chaenomelis Fructus pieces.
