邓老御风膏对高尿酸血症小鼠的降尿酸作用及对尿酸钠致大鼠足肿胀的影响

Effect of Denglao Yufeng Paste on Reducing Uric Acid in Hyperuricemia Mice and Foot Swelling in Rats Induced by Sodium Urate

摘要 目的 探讨邓老御风膏对高尿酸血症(HUA)小鼠的降尿酸(UA)作用及对尿酸钠(MSU)致大鼠足肿胀的影响。方法 将72只KM小鼠和60只SD大鼠分别随机分为对照组,模型组,御风膏低、中、高剂量组,别嘌醇片组。小鼠对照组、模型组给予等剂量0.9%氯化钠注射液灌胃,邓老御风膏低、中、高剂量组分别灌胃给予16.52 g生药/kg、33.04g生药/kg、66.08 g生药/kg,别嘌醇片组灌胃给予45 mg/kg别嘌醇,1次/d,连续7 d。7 d后采用腹腔注射次黄嘌呤建立小鼠HUA模型,对小鼠血清UA进行检测。大鼠对照组、模型组灌胃给予等剂量0.9%氯化钠注射液,邓老御风膏低、中、高剂量组分别灌胃给予11.46 g生药/kg、22.92 g生药/kg、45.84 g生药/kg,别嘌醇片组灌胃给予31.25 mg/kg别嘌醇,1次/d,连续14 d。14 d后于大鼠右足跖皮下注射SUA混悬液造足肿胀模型,于致炎前后测定右足跖容积,计算出注射后1 h、2 h、3 h、5 h、7 h不同时间足跖肿胀度,同时7 h后,取右足跖肿胀组织检测前列腺素E2(PGE2)水平。结果 与模型组比较,邓老御风膏低、中、高剂量和别嘌醇片小鼠UA水平显著降低(P<0.05)。与对照组比较,在注射MSU 1~7 h模型组大鼠足跖肿胀度显著升高(P<0.05);与模型组比较,在注射MSU后1h邓老御风膏低剂量能显著减轻大鼠足跖肿胀度(P<0.05),在注射MSU后1h、2h、5h、7h邓老御风膏中剂量能显著减轻大鼠足跖肿胀度(P<0.05),在注射MSU后7h邓老御风膏高剂量能显著减轻大鼠足跖肿胀度(P<0.05),在注射MSU后7h别嘌醇片能显著减轻大鼠足跖肿胀度(P<0.05)。与模型组比较,邓老御风膏低、中、高剂量组及别嘌醇片组大鼠右足跖肿胀组织中PGE2水平显著降低(P<0.05)。结论 邓老御风膏具有一定的降低UA,缓解SUA所致炎症反应和痛风的作用。 Objective To explore the effect of Denglaoyufeng Paste on lowering uric acid(UA) in hyperuricemia(MSU) mice and foot swelling in rats induced by sodium urate(SUA).Methods 72 KM mice and 60 SD rats were randomly divided into control group,model group,low,medium and high dose groups of Yufeng paste and allopurinol tablet group.Mice in control group and model group were given 0.9% sodium chloride injection by intragastric administration,Deng Lao-yufeng paste low,medium and high dose groups were given 16.52 g crude drug/kg,33.04 g crude drug/kg and 66.08 g crude drug/kg by intragastric administration,respectively,allopurinol tablet group was given 45 mg/kg allopurinol by intragastric administration,once a day for 7consecutive days.7 days later,mice HUA model was established by intraperitoneal injection of hypoxanthine,and serum uric acid was detected.Rats in control group and model group were given equal dose 0.9% sodium chloride injection by intragastric administration,Denglao-yufengpaste low,medium and high dose groups were given 11.46 g crude drug/kg,22.92 g crude drug/kg and 45.84 g crude drug/kg by intragastric administration,respectively,allopurinol tablet group was given 31.25 mg/kg allopurinol by intragastric administration,once a day.14 consecutive days.After 14 days,SUA suspension was injected subcutaneously into the right plantar of rats to create a swelling model of the foot.The volume of the right plantar was measured before and after inflammation,and the swelling degree of the foot was calculated at different times 1 h,2 h,3 h,5 h and 7 h after injection.At the same time,the swelling tissue of the right plantar was taken to detect the level of prostaglandin E2(PGE2) after 7 h.Results Compared with model group,the UA levels of Denglaoyufeng paste low,medium,high dose and allopurinol tablets were significantly decreased(P<0.05).Compared with the control group,the swelling degree of the foot of the model group was significantly increased from 1 to 7 h after MSU injection(P<0.05).Compared with the model group,the low dose of Denglao Yufeng paste at 1 h after MSU injection could significantly reduce the swelling degree of rats' feet(P<0.05),and the medium dose of Denglao Yufeng paste at 1 h,2 h,5 h and 7 h after MSU injection could significantly reduce the swelling degree of rats' feet(P<0.05).High dose of Denglaoyufeng paste could significantly reduce the swelling degree of rats' feet 7 h after injection of sodium urate(P<0.05),and allopurinol tablets could significantly reduce the swelling degree of rats' feet 7 h after injection of sodium urate(P<0.05).Compared with model group,the level of PGE2 in the swollen tissue of right plantar of Denglaoyufeng paste low,medium and high dose groups and allopurinol tablet group was significantly decreased(P<0.05).Conclusion Denglao Yufeng paste has the effect of reducing UA,relieving SUA-induced inflammation and gout.
