两株促进植物生长菌及其混合菌剂对888速生白菜的促生长作用

摘要 为了寻找对作物有促生高效的混合菌剂,本研究利用从广西石角红树林筛选的两株菌株(L113和L35)建立以糖蜜废弃物为载体的混合菌剂,采用钼锑抗比色试验筛选最佳混合菌株配方;通过正交试验确定复合菌株的最佳发酵条件;通过盆栽实验对混合菌株对888速生白菜的促生作用进行验证。钼锑抗实验结果表明,菌株L113和L35配比为1∶1(体积比)、接种量为3%(体积比)进行配制的混合菌株的溶磷量为89.12 mg/L,此条件下配制的混合菌株的磷含量最佳。正交试验结果表明,配制的混合菌株最佳发酵条件为温度30℃,p H值为7.5,发酵时间为72 h,实验验证复合菌株的混菌数为1×109CFU/mL。盆栽试验结果表明,菌株L113和L35混合对白菜具有良好的促生效果,施用混合菌液白菜幼苗的株高、鲜重、干重和叶片数分别较清水对照(CK1)显著提高19.35%、24.22%、33.21%和28.60%,分别较无菌培养基对照(CK2)提高4.22%、16.50%、22.22%和16.36%,说明通过两株混合菌液配制发酵液培养的白菜生长状况优于对照组。本实验以糖蜜为培养基研制的混合菌剂,为复合菌剂的进一步产业化生产和推广提供了菌株配方和重要的实验数据支持。 In order to find a mixed microbial agent with high growth promoting efficiency for crops,this study used two strains(L113 and L35)selected from the Shijiao mangrove forest in Guangxi to establish a mixed microbial agent using molasses waste as the carrier.The optimal mixed bacterial formula was screened using molybdenum antimony resistance colorimetric test;determine the optimal fermentation conditions for the composite strain through orthogonal experiments;validate the growth promoting effect of mixed strains on 888 fast-growing cabbage through pot experiments.The results of molybdenum antimony resistance experiment showed that when the ratio of strain L113 and L35 was 1∶1(volume ratio),and the inoculation amount was 3%(volume ratio),the mixed strain prepared had a phosphorus solubility of 89.12 mg/L.Under these conditions,the phosphorus content of the mixed strain prepared was the best.The orthogonal test results showed that the optimal fermentation conditions for the prepared mixed strain were temperature 30℃,pH=7.5,and fermentation time of 72 hours.The experimental verification showed that the mixed bacterial count of the composite strain was 1×109 CFU/mL.The results of the pot experiment showed that the mixture of strains L113 and L35 had a good growth promoting effect on Chinese cabbage.The plant height,fresh weight,dry weight,and leaf number of Chinese cabbage seedlings treated with the mixed bacterial liquid were significantly increased by 19.35%,24.22%,33.21%,and 28.60%compared to the clean water control(CK1);and 4.22%,16.50%,22.22%,and 16.36%,respectively,compared to the sterile culture medium control(CK2).This indicates that the growth status of Chinese cabbage cultured with the mixed bacterial liquid is better than the control group.This experiment developed a mixed bacterial agent using molasses as the culture medium,providing strain formula and important experimental data support for the further industrial production and promotion of the composite bacterial agent.
