不同盐度和光强对刺枝紫菜自由丝状体生长的影响

摘要 为探究海南野生刺枝紫菜(Pyropia acanthophora)自由丝状体培养的最适盐度和光强条件,将采自海南文昌铜鼓岭的叶状体诱导获得自由丝状体,打碎为长150~200μm藻段后放于锥形瓶中,在温度20℃、光周期12L∶12D培养箱中培养29 d。观察自由丝状体在30μmol·m^(-2)·s^(-1)光强下5个盐度梯度组(15、20、25、30、35),和盐度30时6个光强梯度组(20μmol·m^(-2)·s^(-1)、30μmol·m^(-2)·s^(-1)、40μmol·m^(-2)·s^(-1)、50μmol·m^(-2)·s^(-1)、60μmol·m^(-2)·s^(-1)、80μmol·m^(-2)·s^(-1))下的生长状况。结果显示,刺枝紫菜自由丝状体在35盐度下增重最快,日相对生长速率、叶绿素a、类胡萝卜素、藻红蛋白、藻蓝蛋白和可溶性蛋白含量最高;在30μmol·m^(-2)·s^(-1)光强下日相对生长速率最高,叶绿素a、藻红蛋白和藻蓝蛋白含量最多。综上所述,盐度35、光强30μmol·m^(-2)·s^(-1)是刺枝紫菜自由丝状体培养的最适生长条件。 In order to explore the optimal salinity and light intensity conditions for the culture of Py.acanthophora free filaments,we induced free filaments from thallus collected from Tongguling,Wenchang,Hainan Province,and crushed them into 150-200μm segments.They were then placed in conical flasks and cultured for 29 d under the condition of 12L∶12D at 20℃.The growth of free filaments were observed under five salinity gradient groups(15,20,25,30,35)at the illumination intensity of 30μmol·m^(-2)·s^(-1),and six illumination intensity gradient groups(20μmol·m^(-2)·s^(-1),30μmol·m^(-2)·s^(-1),40μmol·m^(-2)·s^(-1),50μmol·m^(-2)·s^(-1),60μmol·m^(-2)·s^(-1),80μmol·m^(-2)·s^(-1))at a salinity of 30.The results showed that the free filaments of Py.acanthophora grew fastest at a salinity of 35,and the relative daily growth rate,chlorophyll a,carotenoids,phycoerythrin,phycocyanin and soluble protein contents were all the highest.Furthermore,the daily relative growth rate was highest under the illumination intensity of 30μmol·m^(-2)·s^(-1),with the highest contents of chlorophyll a,phycoerythrin and phycocyanin.In conclusion,the optimal growth conditions for free filaments of Py.acanthophora are salinity of 35 and illumination intensity of 30μmol·m^(-2)·s^(-1)。
