大花紫薇ב丹红紫叶’紫薇杂交F_(1)代不育株转录组测序

Transcriptome Sequencing of F_(1) Hybrid Sterile Plants of Lagerstroemia speciosa×Lagerstroemia indica‘Ebony Embers’

摘要 为了从分子生物学角度解析大花紫薇(Lagerstroemia speciosa)和‘丹红紫叶’紫薇(Lagerstroemia indica‘Ebony Embers’)杂交F_(1)代不育株‘潇湘华秀’紫薇(Lagerstroemia indica‘Xiaoxiang Huaxiu’)败育的发生机理,明晰‘潇湘华秀’减数分裂时期相关基因的差异表达情况。采取发育异常的‘潇湘华秀’和发育正常的‘丹红紫叶’的小孢子减数分裂期的花蕾,对其进行转录组测序分析。结果表明:两组间对比共得到14656个差异基因,其中有10427个基因上调表达,4229个基因下调表达。KEGG富集分析表明,这些差异基因主要富集的与减数分裂相关的通路为DNA复制、错配修复、核苷酸切除修复、MAPK信号通路等。对有注释信息的差异表达基因进行筛选,共得到11个与减数分裂相关的基因,其中,差异表达基因ASY1、PRD1、DMC1在减数分裂过程中DNA双链断裂(DSB)的形成与修复、同源染色体的联会配对、染色单体的交叉互换(CO)及二价体的形成中发挥重要作用,这些基因在‘潇湘华秀’减数分裂时期中的低表达或是造成其败育的根本原因。 To elucidate the molecular mechanism underlying the male sterility of the Lagerstroemia indica‘Xiaoxiang Huaxiu’(an F 1 hybrid of Lagerstroemia speciosa×Lagerstroemia indica‘Ebony Embers’)from a molecular biology perspective and to clarify the differential gene expression patterns during meiosis in the sterile line L.indica‘Xiaoxiang Huaxiu’,we performed transcriptome sequencing analysis on the meiotic floral buds of the developmentally abnormal L.indica‘Xiaoxiang Huaxiu’and the normal L.indica‘Ebony Embers’.The results showed that a comparative analysis between the two groups identified a total of 14656 differentially expressed genes(DEGs),with 10427 upregulated and 4229 downregulated.KEGG pathway enrichment analysis revealed that these DEGs were primarily enriched in pathways related to meiosis,including DNA replication,mismatch repair,nucleotide excision repair,and MAPK signaling.Further screening of annotated DEGs identified 11 genes associated with meiosis,including ASY1,PRD1,and DMC1,which play crucial roles in the formation and repair of DNA double-strand breaks,homologous chromosome pairing,chromatid crossover formation,and bivalent formation during the meiotic process.The downregulation of these key meiotic genes in L.indica‘Xiaoxiang Huaxiu’is likely the fundamental cause of its male sterility.
