摘要
目的通过两样本孟德尔随机化(MR)方法探究男性不育与前列腺癌之间的因果关系。方法基于男性不育与前列腺癌的全基因组关联研究(GWAS)资料,通过逆方差加权法(IVW)、加权中位数法(WM)、MR-Egger回归进行MR分析,并通过MR-Egger回归截距项检验、Cochran′s Q检验及留一法评估结果的异质性及水平多效性。结果IVW模型显示男性不育与前列腺癌发病风险无因果关联,MR-Egger回归截距项检验、Cochran′s Q检验及留一法未检测到异质性及水平多效性。结论本研究未能发现明确的证据支持男性不育和前列腺癌发病风险的因果关联,但仍应警惕男性不育对高级别前列腺癌的影响,需要对男性不育与前列腺癌的关系进行更多的研究,尤其是开展针对高级别前列腺癌的研究。
Objective To investigate the causal relationship between male infertility and prostate cancer by two-sample Mendelian randomization(MR)method.Methods Based on the genome-wide association study(GWAS)data for male infertility and prostate cancer,MR analysis was performed using inverse variance weighting(IVW),the weighted median method(WM),and MR-Egger regression.The heterogeneity and horizontal pleiotropy of the results were assessed through the MR-Egger regression intercept test,Cochran′s Q test,and the leave-one-out method.Results The IVW model indicated no causal relationship between male infertility and the risk of prostate cancer.No heterogeneity or horizontal pleiotropy was detected by the MR-Egger regression intercept test,Cochran′s Q test,or the leave-one-out method.Conclusions This study found no clear evidence supporting a causal relationship between male infertility and the risk of prostate cancer.However,attention should still be paid to the impact of male infertility on high-grade prostate cancer.Further research is needed on the relationship between male infertility and prostate cancer,especially for high-grade prostate cancer.
