响应面优化低共熔溶剂提取覆盆子多糖及其抗小鼠运动疲劳作用

Response surface optimization of raspberry polysaccharides extraction using deep eutectic solvents and their anti-fatigue effects on mice

摘要 采用超声辅助低共熔溶剂法提取覆盆子多糖,并进一步研究其抗小鼠运动疲劳作用。对8种不同的低共熔溶剂组合进行了对比筛选,采用响应面法优化工艺获得覆盆子多糖最佳的提取条件,结果为:氯化胆碱-1,4-丁二醇摩尔比1∶4、含水量30%、提取功率140 W、提取温度60℃、提取时间40 min和液固比40 mL/g,在此条件下,覆盆子多糖提取率为90.39%±0.53%,提取物多糖含量为73.15%。建立小鼠负重游泳模型进行抗运动疲劳试验,结果表明:覆盆子多糖能够有效延长小鼠被迫游泳力竭时长;降低运动后小鼠血乳酸、血尿素氮和丙二醛含量;提高超氧化物歧化酶和谷胱甘肽过氧化物酶的含量,平衡小鼠机体的抗氧化作用;增大肌糖原和肝糖原的存量,提升小鼠运动耐力,缓解运动疲劳。 Ultrasonic-assisted extraction of polysaccharides from raspberry using deep eutectic solvents(DES)was investigated and anti-fatigue effects of the extracts were evaluated.A comparison and screening of eight different DES combinations were conducted,the optimal raspberry polysaccharides extraction conditions were determined by response surface methodology.The best conditions for extracting raspberry polysaccharides were:molar ratio of choline chloride to 1,4-butanediol of 1∶4,water content of 30%,extraction power of 140 W,extraction temperature of 60℃,extraction time of 40 minutes,and liquid-to-solid ratio of 40 mL/g.Under these conditions,the extraction yield of raspberry polysaccharides was 90.39%±0.53%,with a polysaccharide content of 73.15%in the extract.An anti-fatigue evaluation was conducted using a weight-bearing swimming mice model.The raspberry polysaccharides significantly prolonged the duration of forced swimming exhaustion in mice,lowered postexercise levels of blood lactate,blood urea nitrogen and malondialdehyde,and enhanced superoxide dismutase and glutathione peroxidase levels,thereby balancing mice body's antioxidant capacity.Additionally,there was an increase in muscle and liver glycogen storage,which contributed to enhanced exercise endurance and fatigue mitigation in mice.
