摘要
目的:利用斑马鱼模型,研究发酵大豆蛋白益生菌复合粉(Gastro-AD大豆蛋白粉和约氏乳杆菌LJ88复配)对幽门螺旋杆菌(Helicobacter pylori,Hp)的抑制作用。方法:通过给予4 dpf斑马鱼不同质量浓度的发酵大豆蛋白益生菌复合粉来确定发酵大豆蛋白益生菌复合粉对斑马鱼感染Hp抑制作用评价的最大检测浓度。以最大检测浓度的1/8、1/4、1/2和1设定4个质量浓度组进行发酵大豆蛋白益生菌复合粉对Hp抑制作用评价及肠道组织结构影响评价。结果:发酵大豆蛋白益生菌复合粉对斑马鱼感染Hp的抑制作用评价的最大检测浓度为1000μg·mL^(-1)。发酵大豆蛋白益生菌复合粉4个质量浓度组对Hp抑制率分别为45.38%(p<0.01)、50.37%(p<0.001)、64.35%(p<0.001)、64.92%(p<0.001),且优于阳性对照组;发酵大豆蛋白益生菌复合粉4个质量浓度组斑马鱼肠腔缩小明显恢复,肠道褶皱数量、肠道组织细胞形态均与正常对照组相似,并随质量浓度上升其作用增强,且优于阳性对照组。结论:Gastro-AD大豆蛋白粉和约氏乳杆菌LJ88组成的复合粉对幽门螺旋杆菌具有明显的抑制作用。
Objective:To study the inhibitory effect of fermented soy protein probiotic compound powder composed of Gastro-AD soy protein powder and Lactobacillus johnsonii LJ88 on Helicobacter pylori(Hp)using zebrafish model.Method:The maximum detection concentration for evaluating the inhibition effect of fermented soy protein probiotic compound powder on zebrafish infection with Hp was determined by administering different concentrations of the compound powder to 4 dpf zebrafish.Then,four concentration groups were set according to 1/8,1/4,1/2 and 1 of the maximum detection concentration to evaluate the effect of fermented soy protein probiotic compound powder on Hp inhibition and intestinal tissue structure.Result:The maximum concentration of fermented soy protein probiotic compound powder for evaluating the inhibitory effect on zebrafish infection with Hp was determined to be 1000μg·mL^(-1).The inhibition rates of Hp in the four concentration groups were 45.38%(p<0.01),50.37%(p<0.001),64.35%(p<0.001)and 64.92%(p<0.001),respectively,which were better than the positive control group.The intestinal cavity shrinkage of zebrafish in the four concentration groups of fermented soy protein probiotic compound powder was significantly restored;the number of intestinal folds and the morphology of intestinal tissue cells in the four concentration groups were similar to those in the normal control group and better than that in the positive control group,with the efficacy enhancing as the concentration increases.Conclusion:The compound powder composed of Gastro-AD soy protein powder and Lactobacillus johnsonii LJ88 had significant inhibitory effects on Helicobacter pylori.
