不同保存温度和时间对血小板聚集结果影响的研究

摘要 目的探究血液标本在不同保存温度和时间条件下,采用光学比浊法测定血小板聚集率的差别。方法收集2023年在该院行血小板聚集检测的健康人群180例,每例采集空腹静脉血两份,共计360份标本,每例其中1份设为对照组置于室温(4 h内完成检测),另外1份设为实验组。将360份标本分为6组,每组60份标本。1组:30份对照置于室温保存,对应30份置于4℃冰箱保存30 min;2组:30份对照置于室温保存,对应30份置于4℃冰箱保存60 min;3组:30份对照置于室温保存,对应30份置于37℃水浴箱保存30 min;4组:30份对照置于室温保存,对应30份置于37℃水浴箱保存60 min;5组:30份对照置于室温保存,对应30份置于-30℃冰箱保存24 h后37℃水浴箱融化5 min;6组:30份对照置于室温保存,对应30份置于-80℃冰箱保存24 h后37℃水浴箱融化5 min,分别检测对照组和实验组花生四烯酸(AA)和二磷酸腺苷(ADP)诱导的血小板聚集率。结果标本放置4℃冰箱不同时刻(30~60 min)PAgT-AA、PAgT-ADP结果与室温相比明显升高,差异有统计学意义(P<0.05);标本放置37℃水浴箱不同时刻(30~60 min)PAgT-AA、PAgT-ADP结果与室温相比差异无统计学意义(P>0.05);标本放置-30℃冰箱及-80℃冰箱24 h后PAgT-AA、PAgTADP结果与室温相比显著降低(P<0.01)。结论不同保存温度和时间会对LTA血小板聚集结果产生不同影响,临床上采集血小板聚集标本后适宜放置室温(20~25℃)保存。 Objective To explore the effect of blood samples on platelet aggregation by light transmission aggregometry under different storage temperature and time.Methods Collected 180 healthy people who at-tended platelet aggregation test in Hebei Petro China Central Hospital hospital in 2023,two samples of fasting venous blood were collected from each case,a total of 360 samples,one sample was set as the control group at room temperature from each case(completing the test within 4 hours),and the other sample was set as the ex-perimental group.360 samples were divided into 6 groups,60 samples for each group.Group1:30 controls were stored at room temperature,and the other 30 samples were stored in 4℃refrigerator for 30 min;Group 2:30 controls were stored at room temperature,and the other 30 samples were stored in 4℃refrigerator for 60min;Group 3:30 controls were stored at room temperature,and the other 30 samples were stored in 37℃water bath for 30 min;Group4:30 controls were stored at room temperature,and the other 30 samples were stored in 37℃water bath for 60min;Group 5:30 controls were stored at room temperature,and the other 30 samples were stored in-30℃refrigerator for 24 hours,and then melted in 37℃water bath for 5 minutes;Group 6:30 controls were stored at room temperature,and the other 30 samples were stored in-80℃refrig-erator for 24 hours and then melted in 37℃water bath for 5 minutes;The platelet aggregation rates induced by arachidonic acid(AA)and adenosine diphosphate(ADP)were measured in the control group and the ex-perimental group.Results The LTA samples were placed in 4℃refrigerator at different times(30-60 min),The results of PAgT-AA、PagT-ADP were significantly higher than that of room temperature,and the differ-ences were statistically significant(P<0.05);the samples were placed in 37℃water bath at different times(30-60 min),the results of PAgT-AA、PagT-ADP were not significantly different from that of room temper-ature(P>0.05);the samples were placed in-30℃refrigerator and-80℃refrigerator 24 hours,the re-sults of PAgT-AA、PagT-ADP were significantly lower than that of room temperature(P<0.01).Conclusion Different storage temperature and time have different effects on platelet aggregation by LTA,it's suitable that placing platelet aggregation samples at room temperature(20℃-25℃)after collection.
