甘露寡糖上调PIgR增强肉鸡肠道免疫屏障

摘要 为了探究甘露寡糖对肉鸡肠道屏障的影响,试验选用1日龄健康白羽肉仔鸡540只,肉鸡饲养至29日龄,按体重相近,公母各半的原则随机分为3组:模型对照组、甘露寡糖保健组和乳酸环丙沙星保健组,每组6个重复,每个重复30只,试验期为10 d。肉鸡30日龄时,模型对照组、甘露寡糖保健组、乳酸环丙沙星保健组肉鸡采用胸肌注射8.37×10^(9) CFU/mL的大肠杆菌菌液0.2 mL进行攻毒,之后分别给予饮用水、0.05 g/L甘露寡糖溶液和60 mg/L乳酸环丙沙星自由饮用,分别连用10 d。试验结束后对肉鸡称重,采集血液测定血清生化指标,采取小肠各段测定器官指数,通过RT-qPCR检测肉鸡肠道屏障关键基因表达。结果表明,模型对照组、甘露寡糖保健组和乳酸环丙沙星保健各组间各器官重、器官指数均无显著差异(P>0.05)。与模型对照组和乳酸环丙沙星保健组相比,甘露寡糖保健组肉鸡血清γ-谷氨酰转肽酶(γ-GGT)含量显著升高(P<0.05)。与模型对照组相比,甘露寡糖保健组肉鸡十二指肠多聚免疫球蛋白受体(PIgR)表达量显著升高(P<0.05);与乳酸环丙沙星保健组相比,甘露寡糖保健组肉鸡空肠PIgR表达量有升高的趋势(P=0.077)。综上所述,在饮水中添加甘露寡糖增加了血清γ-GGT,上调肉鸡十二指肠、空肠免疫屏障PIgR表达,增强肉鸡肠道免疫屏障,维护肠道健康,从而提高机体免疫能力,并相比乳酸环丙沙星效果更为显著。 In order to investigate the effect of mannan oligosaccharides on the intestinal barrier of broil-ers,a total of 5401-day-old healthy white-feathered broilers were selected for the experiment.Broilers were raised to 29 days old and randomly divided into three groups based on the principle of similar body weight with half male and half female:model control group,MOS health group,and ciprofloxacin lactate health group.Each group had 6 replicates,with 30 replicates each,and the experimental period was 10 d.At the age of 30 d,broilers in the model control group,MOS health group,and ciprofloxacin lactate health group were injected into the pectoral muscle with 8.37×10^(9) CFU/mL Escherichia coli bacterial so-lution was used for disinfection.Afterwards,drink-ing water,0.05 g/L MOS solution,and 60 mg/L ciprofloxacin lactate were administered separately for 10 days.Weigh the broilers after all experi-ments are completed,blood was collected for se-rum biochemical indicators,organ indices were measured from various segments of the small intes-tine,and the expression of key genes in the intes-tinal barrier of broilers was detected by RT-qPCR.The results showed that the body weight and organ index of broilers showed no significant differ-ences among the model control group,MOS health group,and ciprofloxacin lactate health group(P>0.05).Compared with the model control group and the ciprofloxacin lactate health group,the MOS health group broiler serumγ-GGT content significantly increased(P<0.05).Compared with the model control group,the MOS health group showed an increase in PIgR in the duodenum of broiler chickens(P<0.05);com-pared with the ciprofloxacin lactate health group,the MOS health group showed an increasing trend in the expression of PIgR in the jejunum of broiler chickens(P=0.077).In summary,adding MOS to drinking water increases serum levelsγ-GGT upregulates the expression of PIgR in the immune barrier of the duo-denum and jejunum of broiler chickens,enhances the intestinal immune barrier of broiler chickens,main-tains intestinal health,and thereby improves the immune capacity of the body,with a more significant ef-fect compared to ciprofloxacin lactate.
