秦艽根和玉米须提取物对高尿酸血症大鼠的治疗效果研究

Study on the Therapeutic Effect of the Extract of the Root of Gentiana macrophylla and Stigmata Maydis on Hyperuricemia Rats

摘要 目的:探究秦艽根和玉米须醇提取物对高尿酸血症(Hyperuricemia,HUA)大鼠的临床治疗效果。方法:制备秦艽根和玉米须乙醇提取物,对Wister大鼠进行干预构建HUA模型,同时采用非布司他片对HUA大鼠进行治疗。通过检测大鼠治疗前后血清中的尿酸(Serum Uric Acid,SUA)、血肌酐(Serum Creatinine,SCre)含量,以及制作HUA大鼠双肾切片,观察其对肾脏组织结构的影响。结果:与模型组相比,高剂量[2.7 mg/(kg·d)]秦艽根和玉米须醇提取物治疗组的大鼠血清中SUA、SCre含量显著降低。结合组织结构观察,发现秦艽根和玉米须醇提取物能有效减轻肾脏肿胀与肾细胞变性坏死情况。结论:秦艽根、玉米须醇提取物可以对HUA大鼠的肾脏起到保护和干预治疗作用,具有一定的开发应用潜力。 Objective:To investigate the therapeutic effect of the ethanol extracts of root of Gentiana macrophylla and stigmata maydis on hyperuricemia(HUA)in rats.Methods:Ethanol extracts of the root of Gentiana macrophylla and stigmata maydis are prepared,and the HUA model is constructed by intervention in Wister rats,and febuxostat tablets are used to treat HUA rats.The serum uric acid(SUA)and serum creatinine(SCre)levels in the serum before and after treatment are detected,and the double kidney sections of HUA rats are prepared to observe their effects on the structure of kidney tissue.Results:Compared with the model group,the serum levels of SUA and SCre in the high-dose[2.7 mg/(kg·d)]ethanol extracts of the root of Gentiana macrophylla andstigmata maydis treatment groups are significantly reduced.Combined with the observation of tissue structure,it is found that the ethanol extracts of the root of Gentiana macrophylla andstigmata maydis can effectively reduce kidney swelling and renal cell degeneration and necrosis.Conclusion:The ethanol extracts of the root of Gentiana macrophylla andstigmata maydis can achieve renal protection and intervention in rats with hyperuricemia from different perspectives,and has certain potential for development and application.
