PGC-1α、HIF1A在胃癌中的表达与临床病理特征及预后的关系

Correlation of PGC-1αand HIF1A expression with the clinicopathological features and prognosis in gastric cancer

目的探讨胃癌组织中过氧化物酶体增殖物激活受体γ辅激活因子1α(peroxisome proliferator-activated receptorγcoactivator-1α,PGC-1α)和缺氧诱导因子1亚基α(hypoxia inducible factor 1 subunit alpha,HIF1A)的表达与临床病理特征及预后的关系。方法选取2017年1月至2021年12月于南京大学医学院附属苏州医院接受手术治疗的110例胃癌患者为研究对象,免疫组织化学法检测胃癌组织中PGC-1α和HIF1A的表达,分析其与临床病理特征及预后的关系。结果PGC-1α表达与TNM分期、浸润深度、淋巴结转移呈负相关(P均<0.05);HIF1A表达与肿瘤直径、分化程度、TNM分期、浸润深度、淋巴结转移呈正相关(P均<0.05)。在胃癌中PGC-1α与HIF1A的表达呈负相关(r=-0.282,P<0.05)。单因素生存分析表明HIF1A的表达与患者生存时间相关(P<0.05)。结论PGC-1α和HIF1A均与胃癌的多个病理指标相关;PGC-1α和HIF1A可能共同作用于胃癌的发生、进展,其机制仍有待进一步研究。

Objective To investigate the expression of peroxisome proliferator-activated receptorγcoactivator-1α(PGC-1α)and hypoxia inducible factor 1 subunit alpha(HIF1A)in gastric cancer tissues and to explore its relationship with clinicopathological features and prognosis.Methods A total of 110 patients with gastric cancer received surgical treatment in the Affiliated Suzhou Hospital of Nanjing University Medical School from Jan.2017 to Dec.2021 were retrospectively enrolled.The expression of PGC-1αand HIF1A was detected by immunohistochemistry,and the relationship with clinicopathological features and prognosis was analyzed.Results The expression of PGC-1αwas negatively correlated with TNM stage,tumor invasion,and lymph node metastasis(all P<0.05).While the expression of HIF1A was positively correlated with tumor size,differentiation,TNM stage,tumor invasion,and lymph node metastasis(all P<0.05).A negative correlation was found between the expression of PGC-1αand HIF1A in gastric cancer(r=-0.282,P<0.05).Univariate analysis showed that overall survival was affected by HIF1A(P<0.05).Conclusion The expression levels of PGC-1αand HIF1A were related to multiple pathological indicators of gastric cancer.The occurrence and progression of gastric cancer may related by the expression level of PGC-1αand HIF1A,and the mechanism needs further investigation.
